Viruses replicate only in living cells so the first viral vaccines were necessarily made in animals smallpox vaccine in the dermis of calves and sheep and rabies vaccines in the spinal cords of rabbits and the brains of mice. Such methods are no longer used in advanced vaccine production and the only intact animal hosts that are used are embryonated hens eggs. Almost all of the vims that is needed for viral vaccine production is obtained from cell cultures infected with vims of the appropriate strain. [Pg.309]

Cell cultures provide infeeted fluids that eontain little debris and can generally be satisfactorily clarified by filtration. Beeause most viral vaccines made fiom cell cultures consist of live attenuated vims, there is no inaetivation stage in their manufacture. There are, however, two important exeeptions inaetivated poliomyelitis vims vaccine is inactivated with dilute formalin or /3-propiolaotone and rabies vaccine is inactivated with /3-propiolactone. The preparation of these inaetivated vaccines also involves a concentration stage, by adsorption and elution of the vims in the case of poliomyelitis vaccine and by ultrafiltration in the case of rabies vaceine. When processing is complete the bulk materials may be stored until needed for blending into final vaccine. Because of the lability of many vimses, however, it is necessary to store most purified materials at temperatures of-70°C. [Pg.309]

The conventional rabies vaccine was administered to the nine initially naive subjects seven days after completing the oral vaccination. Three of these volunteers produced neutralizing antibodies against rabies virus, although none of the five control subjects did. This study showed a clear indication that the orally delivered rabies vaccine has potential as an oral booster for the conventional rabies vaccine. [Pg.154]

The most commonly used screening method for HIV is an enzyme-linked immunosorbent assay, which detects antibodies against HIV-1 and is both highly sensitive and specific. False positives can occur in multiparous women in recent recipients of hepatitis B, HIV, influenza, or rabies vaccine following multiple blood transfusions and in those with liver disease or renal failure, or undergoing chronic hemodialysis. False negatives may occur if the patient is newly infected and the test is performed before antibody production is adequate. The minimum time to develop antibodies is 3 to 4 weeks from initial exposure. [Pg.450]

Viral vaccines are cultivated on inanimate media. Some examples include hepatitis b vaccine, influenza virus vaccine, measles virus vaccine, rabies vaccine, rubella vaccine, and yellow fever vaccine. The viral vaccines are available as lyophilized powder for reconstitution, or suspension for injections,... [Pg.298]

The strength of certain viral vaccines is expressed as international units (IU), which are based on animal potency tests (e.g., rabies vaccine). [Pg.299]

Hospital employees, caring for a patient with rabies, were given 14 days of daily injections followed by booster shots on days 10 and 20 after the fourteenth injection of duck-embryo derived rabies vaccine (DEV). Rabies antibodies first appeared in the serum on day 6 of the injection course, and antibody titers fell significantly after the serum... [Pg.223]

Commonly known as treatment for dog bite. Rabies is usually caused by the bite of infected dog, monkey, cat, etc. and can lead to hydrophobia (feeling of fear of water) and death. A series of five injections need to be given. Usually rabies vaccine is given once the dog bite has already taken place. [Pg.443]
