The concept of the hybrid electrode was extended to a vitamin-immobilized system by Hisaeda and co-workers (Fig. 5), in which heptapropyl and heptaoctyl esters of vitamin B12 derivatives with a Co(II) or Co(III) center were used. Investigation of ti-A isotherms of these compounds with the alkoxysilane monolayers suggested that stable accommodation of the vitamin functionality in the lipid monolayer can be achieved by the introduction of long chains at the vitamin B core. The mixed monolayers of the long chain-substituted vitamin B12 and organosilane amphiphile were transferred as an x-type LB film onto a quartz plate and showed reasonable UV absorbance. The use of an immobilized biofilm system is recommended, since the oxidation products often are only slowly biodegradable. Biomass, especially slow growing microorganisms, can efficiently be kept in the system using support material. This is especially important in continuously operated systems where the liquid phase is completely mixed. In such a system suspended biomass would be washed out when the hydraulic retention time is less than or equal to the population doubling time (i. e. the reciprocal of the specific growth rate Grady, 1985). Efficient immobilization of aptamers on surfaces is necessary for the construction of tongh, stable sensors and assay systems as one necessary step to overcome limitations for practical applications (Bini et al., 2007). Bini et al. (2007) have compared thrombin aptamers immobilized on a gold snrface by chemisorption (thiolated aptamer) and by biotin-streptavidin interaction (biotinylated aptamer carrying a linker) on a gold surface modified by a thiol-dextran-streptavidin layer. The linker-modified aptamer immobilized via streptavidin-biotin showed better reproducibility and sensitivity results for the quartz crystal sensor. Aptamers can be used for the functionalization of titanium-alloy surfaces (e.g., implant material, scaffolds) to enhance cell adhesion. In such hybrid compounds, the nitroxide moiety quenches the fluorescence of the fluorophore (stilbene moiety). The reduction of nitroxide segment by an antioxidant (ascorbic acid) caused a rise in fluorescence of the fluorophore. Mismatched DNA detection was also achieved with FND [35-43]. The affinity of FND for a mismatched DNA area decreases considerably from that for the matched one [38, 41], Therefore, one can estimate the existence of mismatched DNA duplex from a decrease in the current compared with that of the perfectly matched one. Quartz crystal microbalance (QCM) experiments revealed that this current decrease is derived from a difference in the amount of FND concentrated per dsDNA on the electrode. Thus, a thiolated oligonucleotide was immobilized as DNA probe on the gold surface covered on the QCM chip and this chip was dipped in the same electrolyte as the electrochemical measurements. Complementary oligonucleotides with or without a single mismatched base were added in this system. Different lubricants were analyzed by the STM-QCM system, from which some were immobile (iodobenzene on copper) and some were mobile (f-butyl phenyl phosphate on platinum, tricresyl phosphate, and benzene on copper) [61]. Better resolution STM images have been obtained when the quartz of the QCM was oscillating as when the QCM was off for the mobile lubricants, while the quality of the STM image did not change when the QCM was on in the case of the immobile lubricant.
