After obtaining estimates of genome size and restriction-site frequency with pulsed-field electrophoresis, we analyzed 1000 randomly chosen cosmid clones using infrequent restriction sites as landmarks to efficiently determine overlaps, and further clones chosen by cosmid walking. A minimal set of cosmids was restriction-mapped using six enzymes, and where necessary a seventh enzyme was used to determine the exact extent of overlap.

Since overlapping clones failed to provide evidence of the proposed organization of the A locus (Jim Miller, personal communication), definitive mapping of this locus may have to await linkage analysis using pulsed-field electrophoresis [129]. 

Two-Dimensional Pulsed Field Electrophoresis Theoretical Background. Applying the polymer reptation model to conventional DNA electrophoresis is proving to be quite successful, especially in the case of field strength dependent electrophoretic mobilities. 

The silica crystal array can also be used for pulsed fields by adding two additional reservoirs to the PDMS cover to create a 120° crossed field. The mobility of different size long DNA (48.5, 23.1, and 9.4 kbp) vary in the pulsed-field array, indicating that they could be separated in such an array if a suitable injection system were employed. The high electric fields and rapid pulse times employed in these experiments are comparable to pulsed-field electrophoresis in the DNA prism pillar array [37].
