After obtaining estimates of genome size and restriction-site frequency with pulsed-field electrophoresis, we analyzed 1000 randomly chosen cosmid clones using infrequent restriction sites as landmarks to efficiently determine overlaps, and further clones chosen by cosmid walking. A minimal set of cosmids was restriction-mapped using six enzymes, and where necessary a seventh enzyme was used to determine the exact extent of overlap.
