In 2005, the total amount of ephedrine and pseudoephedrine seized in Oceania increased. Ephedrine, smuggled into Australia out of South Africa and Viet Nam, and pseudoephedrine, seized in New Zealand, accounted for most of the total amounts seized. Pharmaceutical preparations were the main source of pseudoephedrine seized at the border and also in clandestine laboratories in Australia and New Zealand. Sizeable amounts of pseudoephedrine tablets, smuggled in a container from Indonesia and by post from Malaysia, were seized in Australia in 2006. In New Zealand, most of the pseudoephedrine seized was in the form of pharmaceutical preparations smuggled into the country out of Asia. Diversions of over-the-counter pharmaceutical preparations and thefts were also reported. The Board recommends to countries and territories in Oceania the launching of appropriate law enforcement activities under Project Prism to further identify smuggling activities.

In view of the methamphetamine problem, the Board encourages Governments to limit, as appropriate, the availability of ephedrine and pseudoephedrine to medical needs, by improving monitoring and control measures over domestic distribution channels, as necessary. In that connection, the Board notes that, in the United States, since the recent initiative of the State of Oklahoma to ban the over-the-counter sale of pseudoephedrine tablets, a number of other states in the United States have now taken steps to monitor the sale of such products.

We proposed using MLC for assay of azithromycin in tablets and capsules. As alternative conventional reversed-phase HPLC method MLC was used for analysis of Biseptol (sulfamethoxazole and trimethoprim) tablets and injection. The MLC was proposed to assay of triprolydine hydrochloride and pseudoephedrine hydrochloride in tablets as alternative normal-phase HPLC method described in USP phamiacopoeia. 

Actifed is a medicinal preparation in which the effective components are the two drugs pseudoephedrine hydrochloride and triprolidine hydrochloride. The absorption spectrum of Actifed tablets dissolved in 0.1M hydrochloric acid is similar to that shown in Fig. 17.14(a) which is clearly of no value for quantitative determinations. A second derivative spectrum however is similar in character to that shown in Fig. 17.14(6) in which peak C corresponds to the pseudoephedrine hydrochloride and D to the triprolidine hydrochloride and from which it is possible to make quantitative measurements. Experience showed that it is advisable to use different response times for the two peaks with the instrument used a response setting of 3 was found to give the best results for pseudoephedrine hydrochloride, whilst a setting of 4 was best for the triprolidine hydrochloride.

Because of their limited effects on allergic symptoms, decongestants often are used in combination with antihistamines.8 Many antihistamines are available in fixed-dose combinations with pseudoephedrine, which enhances the reduction in nasal congestion and allows for the patient convenience of one tablet. Optimally, therapy should be initiated with an antihistamine alone, adding the adrenergic agent only if nasal congestion does not resolve with antihistamine monotherapy. Use of separate antihistamine and pseudoephedrine also permits independent dose titration.4,11,12... 

Pseudoephedrine hydrochloride can be considered a stable compound in bulk and in formulations. After 4-weeks under fluorescent lights (2400 ft. candles) and ultraviolet light (190 pw/cm2) no discoloration or chemical degradation was observed. The bulk drug was stable for at least 6 months at 37°C and 3 months at 50°C. Tablet and syrup formulations stored at 15-30°C for 5 years showed no appreciable degradation. 13... 

An ultraviolet spectrophotometric analysis is used to assay pseudoephedrine hydrochloride in tablets. A portion of finely powdered tablets equivalent to approximately 30 mg of pseudoephedrine hydrochloride is placed in a distilling flask which is part of a micro-steam distillation apparatus. Sodium chloride, water, and concentrated sodium hydroxide are added. A minimum of 30 ml of distillate is collected in a volumetric flask containing dilute hydrochloric acid. The flask is made to volume with distilled water and the absorbance of the solution is determined at 257 nm in 1 cm cells and compared to a solution of known concentration of NF Pseudoephedrine Hydrochloride Reference Standard.1... 

An ultraviolet spectrophotometric method based on the absorbance of a periodate oxidation product of pseudoephedrine hydrochloride will be the official method of analysis in the USP XX.19,20 A portion of tablets or syrup in water is placed in a separatory funnel. Sodium bicarbonate and sodium metaperiodate are added. After standing for 15 minutes, 1 N HC1 is added. The solution is extracted with hexane. The hexane extract is filtered and its absorbance determined at 242 nm in 1 cm cells. The amount of the oxidation product of pseudoephedrine hydrochloride is determined by comparison of the sample absorbance against the absorbance of a Pseudoephedrine Hydrochloride Reference Standard treated in the same manner. 

Commercial samples containing approximately 400 mg of ephedra per capsule yield roughly 5 mg of ephedrine, 1 mg of pseudoephedrine, and less than 1 mg of methylephedrine (White et al. 1997). For a dose of four capsules, yielding approximately 20 mg of ephedrine, the elimination half-life is 5.2 hours. The time to reach maxium concentration is 3.9 hours. Compared to pure ephedrine tablets, the elimination kinetics of ephedra are comparable. However, ephedra showed somewhat different absorption kinetics (e.g., lag time, area under the concentration-time curve, and maximum plasma concentration). So, ephedra tablets may vary from pure ephedrine in the onset of action, but the durations of action are grossly equivalent.

Although initially available as an injectable solution for the treatment of obesity, D-metham-phetamine hydrochloride is currently available as conventional and prolonged release tablets. Illicit methamphetamine is synthesized from the precursors phenylacetone and iV -methylformamide (dl mixture) or alternatively from ephedrine or pseudoephedrine by red phosphorus/acid reduction. 

Since clandestine lab operators use ephedra to make methamphetamine, the DEA monitors supplements containing ephedrine. One kilogram of ephedrine, the equivalent of 48,000 25-mg ephedrine tablets, can be used to produce anywhere from 1.1 to 1.6 lbs (.5 to 0.75 kg) of methamphetamine in a drug lab. Pseudoephedrine can be used for the same purpose.
