Another Fischer achievement was the synthesis of small peptides via the condensation of amino acids. Fischer suggested that there was a common linkage that held pairs of amino acids together in all proteins—the peptide bond. He understood that proteins were tremendously complex, owing to the large number of constituents and the fact of stereoisomerism. By 1916 Fischer had synthesized and characterized 100 peptides, but knew they represented a tiny fraction of what was possible, see also Amino Acid Carbohydrates Isomerism van t Hoff, Jacobus. [Pg.102]

Proteins can also be dangerous or unhealthy. For many who suffer from allergies to agents like pollen, it is proteins on the surface of the pollen that cause an immune response that triggers the allergic reaction. More seriously, many natural toxins are proteins. Biochemistry continues to have a major influence on the development of peptide synthesis. Peptide bond formation via catalysis with proteolytic enzymes has the promise of products with absolute chiral purity and should also be free from many side reactions encountered in synthesis by the methods of organic chemistry. Therefore coupling with the help of enzymes (cf page 57) is receiving growing attention. Perhaps even more exciting is the exploitation of ribosomal protein synthesis for the production of selected target peptides, such as insulin. In recent years preparation of the necessary DNAs became almost routine and hence this avenue of peptide synthesis broadened to a major area that transcends the boundaries of this book. [Pg.99]

Most reactions in cells are carried out by enzymes [1], In many instances the rates of enzyme-catalysed reactions are enhanced by a factor of a million. A significantly large fraction of all known enzymes are proteins which are made from twenty naturally occurring amino acids. The amino acids are linked by peptide bonds to fonn polypeptide chains. The primary sequence of a protein specifies the linear order in which the amino acids are linked. To carry out the catalytic activity the linear sequence has to fold to a well defined tliree-dimensional (3D) stmcture. In cells only a relatively small fraction of proteins require assistance from chaperones (helper proteins) [2]. Even in the complicated cellular environment most proteins fold spontaneously upon synthesis. The ribosome is a ribozyme this is how Cech (2000) commented on the report by Nissen et al. (2000) in Science on the successful proof of ribozyme action in the formation of the peptide bond at the ribosome. It has been known for more than 30 years that in the living cell, the peptidyl transferase activity of the ribosome is responsible for the formation of the peptide bond. This process, which takes place at the large ribosome subunit, is the most important reaction of protein biosynthesis. The determination of the molecular mechanism required more than 20 years of intensive work in several research laboratories. The key components in the ribosomes of all life forms on Earth are almost the same. It thus seems justified to assume that protein synthesis in a (still unknown) common ancestor of all living systems was catalysed by a similarly structured unit. For example, in the case of the bacterium E. coli, the two subunits which form the ribosome consist of 3 rRNA strands and 57 polypeptides. Until the beginning of the 1980s it was considered certain that the formation of the peptide bond at the ribozyme could only be carried out by ri-bosomal proteins. However, doubts were expressed soon after the discovery of the ribozymes, and the possibility of the participation of ribozymes in peptide formation was discussed. [Pg.165]

Upon encountering a stop codon on the mRNA, the ribosome will halt incorporation of further amino acids into the polypeptide as there is no tRNA complementary to a stop codon (UAG, UGA, UAA). In order to liberate the polypeptide, the ester bond between the peptide and the tRNA residing in the P site has to be hydrolyzed — a reaction that is also catalyzed in the peptidyltransferase center. It is critical for protein synthesis that peptide release is tightly coupled to the presence of a stop codon in the decoding center to avoid premature termination resulting in shortened, nonfunctional proteins.
