A reliable analytical method must be available before preformulation studies are started and hence method development activities must precede preformulation activities. The analytical method should be capable of separating the active and any major degradation product(s) and thus be stability-indicating. Analytical methods such as titration and ultraviolet (UV) spectroscopy are not used since they are not considered to be stability-indicating. Only HPLC has been widely used as the method of choice in recent years because of its efficiency, applicability for a wide range of chemical compounds, and ease of [Pg.274]


Several processes progressed to demonstration scales but have not been commercialized, primarily because of economic inabiHty to compete with available petroleum products. The H-Coal process developed by Hydrocarbon Research, Inc. was demonstrated at Catiettsburg, Kentucky using a 545 t/d plant and DOE support. Activated alumina and phosphoric acid on a suitable support have become the choices for an iadustrial process. Ziac oxide with alumina has also been claimed to be a good catalyst. The actual mechanism of dehydration is not known. In iadustrial production, the ethylene yield is 94 to 99% of the theoretical value depending on the processiag scheme. Traces of aldehyde, acids, higher hydrocarbons, and carbon oxides, as well as water, have to be removed. Phase-transfer catalysis is a special type of catalysis. It is based on the addition of an ionic (sometimes non-ionic like PEG400) catalyst to a two-phase system consisting of a combination of aqueous and organic phases. The ionic species bind with the reactant in one phase, forcing transfer of this reactant to the second (reactive) phase in which the reactant is only sparingly soluble without the phase-transfer catalyst (PTC). Its concentration increases because of the transfer, which results in an increased reaction rate. Quaternary amines are effective PTCs. Specialists involved in process development should pay special attention to the problem of removal of phase-transfer catalysts from effluents and the recovery of the catalysts. Solid PTCs could diminish environmental problems. The problem of using solid supported PTCs seems not to have been successfully solved so far, due to relatively small activity and/or due to poor stability. [Pg.8]

HPLC is a very powerful technique for qualitative and quantitative analysis. In the support of process development, HPLC plays an important role in monitoring a reaction, since each reaction component can be quantitated. In this role, the HPLC method must be fast, rugged, and specific, capable of separating all reactants, products, and byproducts.
