Transient reactors, such as pulse (chromatographic) reactors, temporary analysis of products (TAP) reactors, multitrack reactors, and temperature-programmed reactors have been developed mainly to study gas-solid (catalyst) reactions. These are rather sophisticated techniques used to study mechanisms of catalytic processes at the molecular level in great detail. Since this is rarely done in the development of processes for the manufacture of fine chemicals and pharmaceuticals, these reactors are not discussed further. The interested reader is referred to works by Anderson and Pratt (1985) and Kapteijn and Moulijn (1997).

The discovery of BB-3497 was the result of screening a proprietary library for potential metalloenzyme inhibitors at the British Biotech Pharmaceutical Co. Ltd. [73]. This compound was originally prepared in a nonstereoselective manner and its stereochemistry was assigned on the basis of matrix metal-loprotease (MMP) inhibitory activity. The asymmetric synthesis of BB-3497 and the establishment of its stereochemistry by small-molecule X-ray crystallography was later reported by Pratt et al. [83]. Solid-state nmr spectroscopy on a single crystal of KHF2 was interpreted in terms of a centred proton with f = 113.8 pm (Pratt and Smith, 1975). The same conclusion was reached for the other alkali-metal bifluorides (Ludman et al., 1977) and the displacement of the proton from the midpoint was estimated to be less than 60 pm. This technique cannot distinguish between a single minimum and a double minimum with fast transfer. Fluorine-19 nmr spectroscopy has been used to study solutions of HFj" in formic acid, and the data have been interpreted in terms of equilibria involving F , HF, HF2 and H2F2 (Coulombeau, 1977).

Tissue culture has been used to a limited extent for developmental toxicity studies of the studies that have used tissue culture systems, those using chick neural crest cells (Greenberg, 1982) and human embryonic palatal mesenchyme (Pratt et al., 1980, 1982) have been especially useful. In addition, the micromass teratogen test, first described by Flint Orton (1984), has been used as a screening tool, and this test has been used successfully for many years. This method uses cultures of limb and CNS cells. Rat cells are normally used, but mouse and chick cells have also been studied. Many different endpoints can be assessed.
