Peptide research

After the discovery of weak inhibition by the N-terminal products of substrate peptides, researchers at Boehringer Ingelheim reported potent and specific hexapeptides providing up to a 10 -fold increase in potency (Figure 2.3) [89].

Pieter J. Milne was born in Caledon, South Africa in 1955. He completed his undergraduate studies in 1977. After working for 5 years as a pharmacist, he pursued his Masters and doctoral studies under the direction of Dr. L. M. Gerritsma and Professor D. W. Oliver, respectively. In 1994, he was appointed as a lecturer in Pharmaceutical Chemistry at the University of Port Elizabeth, now known as the Nelson Mandela Metropolitan University (NMMU). He became head of the Department of Pharmacy in 2007 and is also the director of the Cyclic Peptide Research Unit (CPRU). 

Thalidomide was initially selected as Candidate K17. It was discovered from peptide research but promoted as being structurally related to barbiturate with hypnotic properties, beyond those of the classic barbiturate Luminal, but without toxicity.

Though naturally occurring peptides based on coded amino acids have been widely used as drugs, there are problems with the use of peptides as therapeutic agents. The problems mainly arise from their rapid metabolism by proteolysis, and their interactions at multiple receptors. The progress of peptide research was expedited by the completion of the Human Genome Project. The availability of DNA sequence information has made it possible to rapidly discover new peptides by screening DNA and expression libraries. Using bioinformatic tools (Antelmann et al., 2001 and Chen et al., 2003), numerous as yet unknown putative polypeptides have been predicted as candidates with secretory properties or as proteases. 

At first glance, blood is the ideal sample from which to obtain a representative clinical sample suitable for peptide research. However, its complexity and the high abundance of certain proteins, such as albumin, fibrinogen, and immunoglobulins, render blood probably the most difficult of all proteomic samples... 

Because of the rapidly developing nature of the field, periodic symposia, such as the one on which this volume is based, will likely proliferate in the next few years in an effort to keep researchers focused on current developments. Such symposia will become more specialized, attending to areas such as sequenced peptides, insect neuropeptide molecular genetics, peptide families, insect neuropeptide processing, and so forth. In this overview, some recent developments in bioactive peptide research are considered, as are directions where such developments may lead.
