Pancreatic tumors antibodies

A combination between MALDI, TLC, and antibody detection was also recently suggested [49] whereby the authors used the following workflow (1) TLC separation of putative cancer-associated GSLs from human hepatocellular and pancreatic tumors, (2) their detection with oligosaccharide-specific proteins, and (3) the in situ MS analysis of the previously protein-(antibody)-detected GSL. Detection limits of less than 1 ng of immunostained GSL could be obtained at these conditions. It is a particular advantage of this approach that only crude lipid extracts of biological sources are required for TLC/IR-M ALDI/MS and no laborious previous GSL purification is needed. [Pg.226]

Mesothelin is a 40-kD glycoprotein of unknown function that is strongly expressed in mesothelial cells, ovarian serous cells, and pancreatic-bile duct cells. Using monoclonal antibody 5B2, Ordonez found it to immu-nostain normal mesothelial cells, mesotheliomas, non-mucinous ovarian carcinomas, and occasionally other neoplasms. Ordonez concluded that mesothelin staining could be used to diagnose mesotheliomas, although it was expressed in 14 of 14 ovarian carcinomas, 12 of 14 pancreatic ductal adenocarcinomas, 7 of 12 desmoplastic small round cell tumors, and 9 of 9 synovial sarcomas. Therefore, this antibody should be interpreted carefully. [Pg.429]

A. Other useful positively staining antibodies include CD56 (NCAM), CD57 (Leu7), and NSE. NESP-55, a member of the chromogranin family, is a promising marker of pancreatic neuroendocrine tumors, but it is... [Pg.519]

With the aid of monoclonal antibodies, sialylated carbohydrate structures, notably sialyl Lea and sialyl Lex, were discovered to function as receptors for the selectins [43]. Sialyl Lex is usually located on leukocytes, but also highly expressed on a variety of different cancer cells [45]. The same holds true for sialyl Lea, which serves as a tumor marker on gastrointestinal and pancreatic cancers [46]. Owing to the function of sialyl Lewis structures in the extravasation of cancer cells from the bloodstream and promoting metastatic spread to other tissues, a clear correlation of expression of sialyl Lea and sialyl Lex on tumors with enhanced progression and metastasis was observed. Since it is assumed that these tumor-associated carbohydrate markers enhance extravasation and metastasis by interactions with selectins, experiments were performed where selectin expression was inhibited. Long-term studies showed that cancer... [Pg.682]
