The Vh/K DNA construct is generated either by RT-PCR (mRNA as template) or PCR (DNA as template). For in vitro transcription/translation, a T7 promoter and protein initiation sequence are added using an upstream primer which also includes degenerate sequences complementary to antibody 5 sequences Table 1), the latter based on Marks et al.
