Numerical Estimates

Make a numerical estimate, with an explanation of the assumptions involved, of the specific surface area that would be found by (a) a rate of dissolving study, (b) Harkins and Jura, who find that at the adsorption of water vapor is 6.5 cm STP/g (and then proceed with a heat of immersion measurement), and (c) a measurement of the permeability to liquid flow through a compacted plug of the powder.

The price of flexibility comes in the difficulty of mathematical manipulation of such distributions. For example, the 3-parameter Weibull distribution is intractable mathematically except by numerical estimation when used in probabilistic calculations. However, it is still regarded as a most valuable distribution (Bompas-Smith, 1973). If an improved estimate for the mean and standard deviation of a set of data is the goal, it has been cited that determining the Weibull parameters and then converting to Normal parameters using suitable transformation equations is recommended (Mischke, 1989). Similar estimates for the mean and standard deviation can be found from any initial distribution type by using the equations given in Appendix IX.

The initial goal of the kinetic analysis is to express k as a function of [H ], pH-independent rate constants, and appropriate acid-base dissociation constants. Then numerical estimates of these constants are obtained. The theoretical pH-rate profile can now be calculated and compared with the experimental curve. A quantitative agreement indicates that the proposed rate equation is consistent with experiment. It is advisable to use other information (such as independently measured dissociation constants) to support the kinetic analysis. 

The kinetic analysis of the sigmoid pH-rate profile will yield numerical estimates of the pH-independent parameters K, k, and k". With these estimates the apparent constant k is calculated using the theoretical equation over the pH range that was explored experimentally. Quantitative agreement between the calculated line and the experimental points indicates that the model is a good one. A further easy, and very pertinent, test is a comparison of the kinetically determined value with the value obtained by conventional methods under the same conditions.

It is experimentally difficult to obtain numerical estimates of the total number of bacteria present in seawater, and the contribution of ultramicroorganisms that have a small cell volume and low concentrations of DNA may be seriously underestimated. Although it is possible to evaluate their contribution to the uptake and mineralization of readily degraded compounds such as amino acids and carbohydrates, it is more difficult to estimate then-potential for degrading xenobiotics at realistic concentrations. 

Finally, the user should always be aware of the danger in getting numerical estimates of the derivatives from the data. Different smoothing cubic splines or polynomials can result in similar values for the state variables and at the same time have widely different estimates of the derivatives. Based on the above expressions, it is obvious that by this approach one needs to estimate numerically the time derivatives of Xv, S and P as a function of time. This can be accomplished by following the same steps as described in Section 7.1.1. Namely, we must first smooth the data by a polynomial fitting and then estimate the derivatives. However, cell culture data are often very noisy and hence, the numerical estimation of time derivatives may be subject to large estimation errors.

Leung 1993). PBPK models for a particular substance require estimates of the chemical substance-specific physicochemical parameters, and species-specific physiological and biological parameters. The numerical estimates of these model parameters are incorporated within a set of differential and algebraic equations that describe the pharmacokinetic processes.
