Incubation periods in excess of 2 h were required before this activity was detected in cell-free supernatants. More recently, the use of cDNA probing of Northern transfers (to detect specific mRNA levels), the use of ELISA techniques (to detect protein levels immunologically) and the development of more specific bioassays (culture techniques in which a biomolecule stimulates proliferation in a particular cell line) have resulted in a more thorough analysis of IL-1 production by neutrophils. IL-1 is only poorly expressed in blood neutrophils because mRNA for this cytokine is detectable only at very low levels (if at all), and protein production is usually below the level of detection of most assays. However, exposure of neutrophils to lipopolysaccharide (LPS), or to cytokines such as GM-CSF, TNF or IL-1 itself, results in a rapid but transient increase in IL-1 expression. [Pg.250]

Liquid solvents in this picture may be considered as islands in the northern hemisphere, because the state of molecules in a liquid solvent is quite definitely closer to the COSMO state than to the vacuum. The only exception may be alkane solvents, which are located somewhere close to the equator due to their fully nonpolar character. Contamination of the biosphere by stable OCPs (DDT, HCH, aldrin, dieldrin, heptachlor, and toxafene) has become a global phenomenon. With global transfer, pesticides may reach countries that never produced or used them. In the northern hemisphere, where winds tend to blow from west to east, the rate of the wind in the boundary layer between the troposphere and the atmosphere is approximately 35 m/sec. Caribou in northern Quebec contained up to 1129 Bq 137Cs/kg muscle FW in 1986/87, but only 10 to 15% of this amount originated from Chernobyl the remainder is attributed to fallout from earlier atmospheric nuclear tests (Crete et al. 1990). The maximum concentration of 137Cs in meat of caribou (Rangifer tarandus granti) from the Alaskan Porcupine herd after the Chernobyl accident did not exceed 232 Bq/kg FW, and this is substantially below the recommended level of 2260 Bq 137Cs/kg FW (Allaye-Chan et al. 1990). Radiocesium transfer in an Alaskan lichen-reindeer-wolf (Canis lupus) food chain has been estimated. Here, instead of DNA fragments, mRNA fragments are probed with a labelled cDNA probe after separation by electrophoresis and transfer to nitrocellulose membranes.
