Murine monoclonals, antigenicity

Muronomab-CD3 is a murine monoclonal antibody directed against the CD3 complex of the T-lymphocyte antigen receptor. This drug selectively diminishes the T-lymphocyte pool resulting in a strong lymphopenia. Similar to other nonhuman antibodies the generation of human antimurine antibodies (HAMA) limits its long-term use. [Pg.619]

Initial stages in the production of murine monoclonals entail administration of the antigen of interest to a mouse. This is followed by sacrifice and recovery of activated B-lymphocytes from the spleen. A similar approach to the production of human monoclonals would be unethical. Administration of some antigens to humans could endanger their health. Although B-lymphocytes could be obtained from the peripheral circulation, the majority of these are unstimulated, and recovery of (stimulated) B-lymphocytes from the spleen is impractical. [Pg.391]

The first monoclonals were murine (mouse) antibodies, because the early experiments involved injecting mice with an antigen, harvesting their B cells, and then fusing a harvested B cell with an immortal tumor cell line. There were immediate ahempts to make therapeutic drugs from murine monoclonal antibodies. Scientists at Harvard Medical School tried to apply murine monoclonal antibodies for heahnenf of tumors as early as 1980. But with a very few exceptions, murine antibodies failed as drugs in humans. [Pg.568]

Described here is an indirect method for detecting two different cellular antigens in acetone-fixed tissue, using a rabbit polyclonal antibody, and a murine monoclonal antibody on the same section. One secondary antispecies antibody is conjugated with alkaline phosphatase, the other with peroxidase, thus resulting in two differently colored products showing the localization of the two antigens (Fig. 1). [Pg.271]
