The mouse lymphoma assay is in fact just one of several mammalian cell assays designed to determine increases in mutation rate. It focuses on the thymidine kinase (tk) assay in murine lymphoma cells (L5178Y), though tk data have also been produced from the human lymphoblastoid cell line TK6, and at the hgprt (hypox-anthine-guanine phosphoribosyl-transferase) locus in Chinese Hamster ovary or lung (V79) cells and mouse lymphoma cells. Since Ames mutation data and often... [Pg.254]

Gs is established to be the stimulatory regulatory component of adenylyl cyclase. This was first demonstrated by its ability to reconstitute the adenylyl cyclase system of cyc cells [3,22] concomitant with the reappearance of CTX labeling [23]. Cyc cells are derived from S49 murine lymphoma cells and lack Gs as indicated by lack of stimulation of adenylyl cyclase by NaF, GTP analogs and hormones (in spite of the presence of stimulatory receptors), by lack of substrate for CTX and by lack of mRNA encoding Gs-a subunits. Moreover, pure Gs also stimulates a resolved C preparation [24], as well as fully purified C [25,26] of adenylyl cyclase, both in solution [27] and after reconstitution into phospholipid vesicles [28]. Thus, by all criteria the purified Gs is functional Gs. [Pg.5]

Some daphniphyllum alkaloids, such as calydphyllines A (82) and B (83), exhibited moderate cytotoxicity against murine lymphoma L1210 cells in vitro [84]. Daphni-glaucin C showed inhibition of polymerization of tubulin at IC50 25mg/mL [82]. Recently, some daphniphyllum alkaloids such as daphmanidins E and F showed moderate vasorelaxant activity on rat aorta [80]. However, since the pharmacological activity of the daphniphyllum alkaloids is poorly studied, this area should be developed in future. [Pg.585]

DS-penaustrosides A (79) and B (80) revealed cytotoxic activities against murine lymphoma L1210 and human epidermoid carcinoma KB cell lines [IC5o=0.12 (L1210) and 0.62 (KB) p.g/ml for 79 2.1 (L1210) and 4.6 (KB) ig/ml for 80] [69]. Unfortunately, the authors did not show the data on activities of relative holostane derivatives in the same experimental conditions. [Pg.158]

In a recent study [55], (III. 156), (III. 160), and the lO-CHO derivative (III. 163) were tested against DHFR from HCT-8 human colon tumour cells, and were found to have IC50 values of 0.05,0.35, and 0.009 pM, respectively, as compared with 0.0013 /rM for MTX and 0.00065 /rM for 5-methyl-5,8-dide-azaAMT (III. 148). An interesting feature of these results was the rather high potency of (III. 163), which was consistent with the data obtained against DHFR from L5178Y murine lymphoma cells by Dedhar et al. [47] and was reminiscent of the anti-DHFR activity reported a number of years ago for A °-formylfolate [56]. [Pg.41]
