This chapter describes the conditions required for successful microinjections. Furthermore, examples of repetitive injections or multiple puncturing of a single giant vesicle are shown. 

One of the main reasons that GVs are so attractive for studying is their use as microreactors. In such systems the progress of enzymatic reactions in a compartmentalized system can be studied in situ and in real time (see also chapter 22). An early example of such a system is the digestion of microinjected DNA by pancreatic DNase I in a GV This system also shows the feasibility of this microinjection technique in GVs, because it demonstrates that multiple puncturing of a target GV is possible. 

Multiple punctures in the iliac crests (more than 100-150) are necessary to aspirate enough marrow cells for transplantation. This procedure is made under general or less frequently regional anesthesia. Harvest of marrow cells from the sternum is exceptional. Marrow blood (1000-1500 ml) has to be obtained and subsequently filtered to eliminate bone fragments. Besides new cases of already known side effects some cases of uveitis and vitiligo after intradermal treatment of malignant melanoma have been described for the first time (13 ). One case of Guillain-Barre syndrome after treatment by a multiple puncture technique has been published (14 ). 

To use the port, the overlying skin is prepared using conventional techniques. A local anesthetic is sometimes used to decrease pain of needle insertion, though this is usually not necessary using techniques which utilize small-bore needles. A special point needle is used to puncture the implanted ports as the point of these needles is deflected so it tears the septum rather than coring it, allowing multiple entries. The septum reseals when the needle is removed.

This chapter introduces workers in various industries to several more hazards they must guard against high pressure, steam, and electricity. Remember we said in Chapter 1 that risk was everywhere Maybe we should have added that multiple risks are everywhere. All work sites have multiple risks, from heights (stairs), electricity (office equipment), punctures (stapler, scissors, etc.), ergonomics, and many more.
