Online detection using 4H nuclear magnetic resonance (NMR) is a detection mode that has become increasingly practical. In a recent application, cell culture supernatant was monitored on-line with 1-dimensional NMR for trehalose, P-D-pyranose, P-D-furanose, succinate, acetate and uridine.33 In stopped-flow mode, column fractions can also be analyzed by 2-D NMR. Implementation of SFC has initially been hampered by instrumental problems, such as back-pressure regulation, need for syringe pumps, consistent flow-rates, pressure and density gradient control, modifier gradient elution, small volume injection (nL), poor reproducibility of injection, and miniaturised detection. These difficulties, which limited sensitivity, precision or reproducibility in industrial applications, were eventually overcome. Another important environmental and forensic application is the identification of explosives and their degradation products. A method based on liquid chromatography was unable to produce baseline resolution of 14 nitroaromatic and nitramine explosive compounds. The problem was solved as Bailey et al. tested the separation of the same mixture in CEC mode (column packed with 1.5-pm C18 silica) [89]. A baseline resolution of all 14 compounds was realized in 7 min in a second run, performed in less than 2 min, only 2 analytes of 14 co-eluted. To stabilize EOF and reduce peak tailing, SDS was added to the mobile phase.
