In recent years h.p.l.c. has become a valuable chromatographic tool for analytical and preparative scale work. In this latter area the separation of isomers (structural, diastereoisomeric, and enantiomeric) has been possible by the selection of appropriate column packing material and solvent systems. However, the equipment, operating costs, and column packing materials are more expensive than those in t.l.c., g.l.c. and conventional liquid-solid column chromatography.

Specific and essentially stand-alone mode of liquid chromatography is associated with the absence or suppression of any analyte interactions with the stationary phase, which is called size-exclusion chromatography (SEC). In SEC the eluent is selected in such a manner that it will suppress any possible analyte interactions with the surface, and the separation of the analyte molecules in this mode is primarily based on their physical dimensions (size). The larger the analyte molecules, the lower the possibility for them to penetrate into the porous space of the column packing material, and consequently the faster they will move through the column. The schematic of this classification is shown in Figure 1-1. 

There are a variety of column packing materials and liquid phases. Sometimes a very long capillary column is used without a solid packing and only coated with a liquid on the wall. The oven can be operated isothermally at a given temperature or according to a program where temperature is varied as a function of time.

Although separation occurs by sieving or exclusion of the constituents by the column packing material, other so-called non-size exclusion effects are also known to occur. The non-size exclusion effects can be divided into two major groups ionic interactions and adsorption. The extent of these interferences is determined in part by the type and process used in the manufacture of the column packing material, and in part by the experimental conditions.

Future development is likely to include improvements in column liquid-liquid chromatography—new column packing materials and new detection systems. Stationary phases consisting of specific groups covalently bound to inert matrices are being developed in many laboratories for use in liquid-liquid as well as gas-liquid chromatography. Such phases might be made for use in bile acid analysis.
