Interferon-7 was the first secretory product of T cells to be discovered when it was found that supernatants derived from suspensions of T cells that had been treated with mitogenic agents could activate macrophages. This macrophage activating factor , subsequently found to interfere with the replication of viruses, was thus named interferon. The production of this compound, associated with delayed-type hypersensitivity and cell-mediated immunity, was termed immune interferon or type II interferon. With the discovery of other lymphokines with interferon-like activity (interferon-a and -j3), the compound was finally designated interferon-7.

IFNy was formerly known as immune IFN, macrophage activating factor, T-ceU IFN, and type II IFN. 

The major biological activity attributed to IFNy is enhanced antigen presentation. This activity is effected by the induction of MHC class II antigens on a variety of cells, including epithelial, endothelial, and connective tissue cells, and monocytes (B32, P16, S51, V7). IFNy also acts as a macrophage activating factor (A6, YIO) and synergizes with TNF to effect tumor cell lysis and inhibit cell proliferation (S6).

This poorly defined factor is called macrophage activating factor (MAF). In contrast certain interferon inducers, such as endotoxins and poly (I)-poly (C) can transform macrophages without the need for lymphocytes.24 Direct activation of macrophages by pyran copolymer and other polyanionic inducers without lymphocyte-derived MAF was also demonstrated.2 28 Since these inducers stimulate macrophages to produce interferon, and interferon activates macrophages, it is reasonable to think that interferon is the effective activator in these instances.
