BicoZamycin. Bico2amycin (bicyclomycin) [38129-37-2] C22H2gN20y, shown in Figure 6, is a colorless, water-soluble diketopipera2ine or cycHc dipeptide derived from leucine and isoleucine. It was simultaneously isolated in 1972 in Japan from cultures of two different strains (166). [Pg.154]

By measuring the L-isoleucine/o-alloisoleucine ratio in the protein isolated from the eggshells of an extinct Australian bird, a team of scientists recently determined that this bird lived approximately 50,000 years ago. Radiocarbon ( " C) dating is not accurate for samples older than about 35,000 years, so AAR is a useful addition to the tools available to paleontologists. [Pg.1116]

The lantibiotic cypemycin isolated from a Streptomyces sxi21tl exhibits bis-methylation at Alai (Me2N-Ala) and an L- //o-isoleucine at position 13. The gene for its precursor peptide has not been reported, and hence the amino acid that is modified to generate the allo-We is not known. If it is introduced posttranslationally like all other modifications in lantibiotics, its most likely precursors would be either He or Leu and the posttranslational modification may involve a radical mechanism as neither amino acid is activated at C/3 or C7 for heterolytic chemistry. [Pg.240]

[67] used Arabidopsis sequences to design primers to clone the missing steps from a cDNA library. They have expanded the schema to include the biosynthesis of the key precursors phenylalanine and leucine, valine and isoleucine. Leucine.—The greater part of the leucine is contained in the ester fractions, which boil between 70° and 90° C. It generally occurs in considerable amounts in the protein, and is obtained by crystallisation from water, in which it is less soluble than the other amino acids which may be present. It is seldom present in its pure, optically active form, as this is easily racemised, and the various crops of crystals most probably also contain isoleucine. It is more easily isolated by completely racemis-ing the mixture of amino acids contained in this fraction by heating in an autoclave with baryta to 160-180° C., and then, after removal of the baryta, separating it by crystallisation. The difficulty of separating it from the other amino acids, especially valine and isoleucine, makes an exact quantitative estimation almost impossible. The values which have been found are therefore minimal ones, and they will also include in many cases the yield of isoleucine. [Pg.12]

From such a background, some kinds of polypeptide blend samples have been studied by solid state NMR.27,72 74 Especially, detailed information for four kinds of blend samples such as poly(L-alanine) (PLA)/poly(L-valine) (PLV), PLA/poly(L-isoleucine) (PLIL), poly(D-alanine) (PDA)/PLV and polyglycine (PG)/PLV blends, have been reported. Here, let us describe some reasons why PLA/PLV, PDA/PLV, PLA/PLIL and PG/PLV blends are interesting systems. PLA and PDA in the solid-state can take the a-helix and (3-sheet forms due to intra- and intermolecular HBs, respectively. PG in the solid-state can take the 3j-helix (PG-II) and (3-sheet (PG-I) forms due to intra- and intermolecular HBs, respectively. However, PLIL and PLV in the solid state can predominantly take the (3-sheet form as the stable conformation. For this reason, it is interesting to know whether an isolated a-helix or 3i-helix form polypeptide surrounded by a major polypeptide in the (3-sheet form can take the helical conformation, or not, due to the balance between intramolecular and intermolecular hydrogen bonds. In addition, we would like to know whether a polypeptide in the (3-sheet form surrounded by a major polypeptide in the a-helix or 3 -helix form can take the (3-sheet form. [Pg.7]

In this section, tetramic acids with an acyl group substituent at C-3 are discussed. The simplest of the naturally occurring 3-acyl tetramic acids, tenuazonic acid (6), was first isolated from the culture filtrate of Alter-naria tenuis [18] and, subsequently, from other fungal species (A. alternate, A. longipes, Pyricularia oryzae) [19,20]. Species of Altemaria are known to produce more than 70 secondary metabolites, many of which, particularly those from the Altemaria altemata complex, are mycotoxins [19]. The absolute stereochemistry of 6 (55,65) was deduced from the formation of L-isoleucine on ozonolysis followed by acid hydrolysis [21]. [Pg.114]

Kirchgessner and Steinhart (52) studied the in vitro digestion of soy protein isolate with pepsin. They showed that a relatively higher percentage of the essential amino acids threonine, valine, isoleucine, leucine and phenylalanine were found in the undigested residue and therefore would be present in lower proportions in the hydrolysate. Myers et al. ( ) hydrolyzed a soy protein isolate with a fungal acid protease. They also found that the essential amino acid content of the hydrolyzate was lower than the isolate, but, as prepared by their continuous process, the hydrolyzate PER was not significantly reduced as compared to the isolate PER. [Pg.254]

Amino Acids. Traces of amino acids were found in the Devonian Newton Hamilton and Marcellus Formations in a previous study (7). In the present work (Table V) small quantities of amino acids were isolated from the Marcellus shale. In both the earlier work and the present study an amino acid that chromatographed as arginine was found. Other amino acids in the Marcellus black shale are histidine( ), methionine, alanine, tyrosine, valine, leucine or isoleucine, and two unknowns, possibly including aminobutyric acid of nonprotein origin in the Newton Hamilton Formation histidine( ). Ammonia was also present in both the Marcellus and the Newton Hamilton. [Pg.17]

Thermolysin is a metalloenzyme isolated from Bacillus thermoproteo-lyticus. It is a heat-stable extracellular endopeptidase of molecular weight 34,600. The enzyme catalyzes the hydrolysis of peptide bonds that have the amino group as part of hydrophobic residues such as phenylalanine, isoleucine, or leucine. [Pg.326]

In 1963 a group of Swiss authors (12) isolated zizyphine from Zizyphus oenoplia and recognized isoleucine and proline as components. Two years later Zbiral etal. (13) proposed a complete structure which was later revised (14). Pais et al. (15) in an earlier preliminary report suggested the structure of pandamine which had been isolated from Panda oleosa. This was confirmed in 1966, and the structure of the similarly constituted pandamine was reported (17). Shortly thereafter Tschesche and co-workers (18) reported the structure of an alkaloid of this type scutianine-A from Scutia buxifolia. Since then the number of cyclopeptide alkaloids of known structure has risen to more than sixty, a figure which Klein and Rapoport had envisioned in 1968 (20). The workers at Gif-sur-Yvette and at Bonn have been in the forefront of these researches but others have made important contributions (19-26). [Pg.166]
