When normal serum immunoglobulin levels are compared (see the table) it can be seen that more sensitive methods are required for the quantification of total IgE than for other immunoglobulins. Clearly, sensitivity will be an even more important characteristic of methods designed to measure allergen-specific IgE antibodies. 

Another hypothesis for the decreased rates of atopy observed among helminth-infected individuals is that parasite-induced polyclonal immunoglobulin E (IgE) compete with allergen-specific IgE for FccRI binding on basophils and mast cells. Recently, however, this was not confirmed (Mitre et al. 2005).

These allergens bind to allergen-specific immunoglobulin E (IgE) on mast cells, crosslinking the IgE molecules and aggregating the underlying FceRI receptors. Activation of mast cells leads to the release of several mediators, like histamine.

Sensitivity of immunoassays is largely conditioned by markers applied for conjugation with the antibody. Traditional immunodetection methods ELISA with radioactive markers (radioimmunoassay—RIA), enzymatic markers (enzyme immunoassays—El A), or fluorescent markers (fluoroenzyme immunoassays FEIA) are currently the most widely used techniques in laboratory analysis of allergens as well as in clinical studies for determination of general and specific IgE and other subclasses of immunoglobulines, e.g., IgG4 in the Immuno-CAP system (Samson, 2001 Duran-Tauleria et al., 2004 Lidholm et al., 2006).
