Fig. 9. (a) H, 15N] HSQC NMR spectra of 15N-labeled 7 and 2 mol equiv of 5 -GMP after lmin and 30 min of irradiation (, 195Pt satellites) (b) time dependent decrease in concentration of 7 and formation of 7rG and 7rG2 (lines drawn merely to connect points) (c) quantification of in vitro DNA repair synthesis using an extract prepared from the repair-proficient HeLa cell line. Cisplatin was taken as 100%. Adapted from Ref. (35). [Pg.18]

It has taken many years, however, for a callus culture to be described which produces vinblastine (1). Success was achieved by Miura and coworkers 122) as a result of the screening of callus tissues with the HeLa cell line. Vinblastine (1) was detected at a level of 1 p,g/g dry weight by HPLC and characterized by retention time and mass spectrometry. This level is below that in the whole plant, and thus its presence was probably overlooked by previous workers. [Pg.42]

Corticotropin (ACTH) receptors have been used as the test preparations for comparison of the binding affinities of adrenocorticotropin peptides using a cloned mouse adrenocorticotropin receptor expressed in a stably transfected HeLa cell line (Kapas et al. 1996). Such assays measure the initial membrane binding but not strictly the biological activation e.g. of adrenal cells or melanocytes. [Pg.353]

Inhibition of cellular prenylation in HeLA cell lines compared to saturated prenylation, measured by reprenylation signal using biotin-GPP. nd, no data. [Pg.199]

They are flavonoids without the C-ring and are not frequently isolated from plants, possibly due to their instability. So, most of the chalcones presented in this review are synthetics [80, 79]. This kind of flavonoids, generally, are rather cytotoxic against human cancer cells. Thus, xantohumol, a prenylated chalcone from hops, inhibited the A-2780 cell line at a IC50 of 0.5 pM [56] and two synthetic chalcones inhibited the HeLa cell line at IC50 of 0.0038 pg/mL [80]. [Pg.926]

The selection of the appropriate cell line is critically important and should be dictated by the goals of the study. The selection is not easy as no cell line is perfect almost no matter what the objective of the study is. More often than not it seems that current conventions of the field, past or current local use, or easy availability are the principal reasons for selecting a certain cell line as a workhorse for particular studies. This may be dangerous as has been amply proven by the example of HeLa cell line. This cell line has been used over several decades for numerous purposes without proper characterization, and furthermore, it has contaminated or replaced other... [Pg.509]

The strain Phomopsis sp. hzlaOl-1 produced two very closely related new 10-membered macrolides, phomolides A (28) and B (29), and one new benzofuran derivative (30). All of them displayed antimicrobial activities against Escherichia coli, Candida albicans, and Saccharomyces cerevisiae, while 30 also showed strong activity against Bacillus subtilis. When tested for cytotoxicity, none of the compounds was active toward the HeLa cell line. [Pg.231]

Zinc(ii) complexes of (jE )-N-(pyridin-2-ylmethylene)arylamines (L) of the general formula [Zn(X)2(L)] (X = Cl , Br , I ) (see Fig. 4) with distorted tetrahedral or square pyramidal coordination geometries have been reported to show a broad fluorescence emission band at Aem = 410nm. Moreover these species exhibited significant cytotoxic activity toward the HeLa cell line and their high IC50 values did not induce any cytolysis effect." ... [Pg.152]

In order to observe the effects of PROTAC-4 on FKBP, a HeLa cell line stably expressing a GFP-tagged FKBP was generated. Thus, loss of the target protein could be visually assessed by diminished fluorescence. The GFP-FKBP cells were then treated with PROTAC-4, which was simply added to the cell... [Pg.78]
