Global Effects

For this experiment, as well as for the microtomography ( 3.2) we used the commercial desktop microtomography system Skyscan 1072 [5], the setup of which is sketched in Figure 1. For this instrument, which is designed to study relatively large objects with a diameter up to 50 mm, the source size is 8 pm, the distance source-detector is about 50 cm and the effective resolution of the detector is about 80 pm. For this system and this object, the global effective resolution a is estimated to be of the order of 50 to 100 pm [6]. 

Failure Cause. The failure cause is the physical, chemical, electrical, thermal, or other design deficiency which caused the failure. The agent, physical process, or hardware deficiency causing the failure mode must be identified, ie, what caused the failure for each failure mode. There may be more than one cause. Failure Fffect. The failure effect is the local effect on the immediate component/subsystem and the global effect on system performance/operation. In commercial products, the effect on the customer, ie, the global effect, must be addressed. 

Atmospheric emissions are controlled by legislation because of their damaging effect on the environment and human health. These effects can be categorized into their local effects and global effects. There are four main problems associated with atmospheric emissions2 3.

In another intriguing study, Rosania showed that chemical substructures can be predictive of subcellular distribution, which could be highly relevant to the current topic of target prediction [82]. 

When 18-crown-6 was co-lyophilized with a-chymotrypsin, a 470-fold activation was seen over the free enzyme in the transesterification of APEE with 1-propanol in cyclohexane (Scheme 3.2) [96]. There was a low apparent specificity for the size and macrocyclic nature of the crown ether additives, suggesting that, during lyophilization, 18-crown-6 protects the overall native conformation and acts as a lyoprotectant. To examine this global effect, FTIR was used to examine the effect of crown ethers on the secondary structure of enzymes. In one study [98], subtilisin Carlsberg was shown to retain its secondary structure in 1,4-dioxane when lyophi-lized in a 1 1 ratio with 18-crown-6. In addition, examination of FTIR spectra from varying incubation temperatures indicated that an increase in crown ether content in the final enzyme preparation resulted in a decreased denaturation temperature in the solvent, indicating a more flexible protein structure.
