In 1991, Kojiri et al. reported the isolation of BE-13793C (344), an alkaloid isomeric with the arcyriaflavins C (347) and D (348) (see Scheme 2.89), from the culture broth of Streptoverticillium moharaense strain BA 13793 collected in Seto, Aichi Perfecture, Japan. BE-13793C (344) showed strong inhibitory activity against topoisomerases I and II and inhibited the growth of doxorubicin-resistant or vincristine-resistant P-388 murine leukemia cell lines, as well as their parent P-388 cell line (330). [Pg.139]

Vaclavikova et al. [216] have investigated the effect of 13 flavonoid derivatives—aurones, chalcones, flavones, flavonols, chromones, and isoflavones—on 14 C-paclitaxel transport in two human breast cancer cell lines, the doxorubicin-resistant NCI/ADR-RES and sensitive MDA-MB-435. The compounds with known binding affinity toward the NBD of P-gp were selected. The four aurones studied most effectively inhibited P-gp-related transport in the resistant fine in comparison with other groups of flavonoids. The aurones also most effectively increased the intracellular accumulation of paclitaxel and decreased its efflux. The results obtained did not always correlate with the binding of flavonoid derivatives to P-gp, so this indicated that the binding was not the only factor influencing the transport of paclitaxel. The different aspects of inhibition of P-gp by polyphenols was recently reviewed by Kitagawa [212],... [Pg.274]

Much earlier, the partitioning of [14C]-doxorubicin was determined in subcellular fractions of doxorubicin-sensitive (V-79) and doxorubicin-resistant (LZ) hamster lung fibroblast cell lines. A significant difference was reported (Table 5.14) [114], At concentrations clinically relevant in vivo, doxorubicin preferentially partitioned into the membrane, achieving a 100-fold higher concentration in the membrane fraction than the cytosol fraction. The data in Table 5.14 show the ability of resistant cells to decrease the doxorubicin content in the lipid fraction of the membrane. It is remarkable that the doxorubicin content of the lipid membrane fraction in resistant cells decreased about ninefold, whereas the reduction in whole cells is only fourfold in com-... [Pg.260]
