DNA adducts are excellent biomarkers in determining the extent of genetic damage. These markers are useful in understanding the carcinogenesis mechanism and the assessment of cancer risk imposed by several chemical agents. Considering the importance... [Pg.981]

The Phase II acetylation of aromatic hydroxyl-amineS/ the products of Phase I metabolism of aromatic amineS/ constitutes a toxic metabolic pathway that has been implicated in carcinogenesis/ as illustrated in Scheme 11.36. Rapid acetylators (with respect to NAT2) have been shown to be associated with an increased risk of colon cancer. The mechanism of this toxicity has implicated the intermediacy of the reactive... [Pg.158]

Relatively minimal information is available on cancers associated with fibre exposure. Lechner etal. (1997) summarised these carcinogenesis mechanisms with primary emphasis on the alterations described in human mesotheliomas. Of the known changes, the most frequent are in the tumour-suppressor genes (Kratzke etal. [Pg.56]

Experimental animal studies have played a key role in the understanding of the mechanisms of chemical carcinogenesis. The duration of development of a cancer in humans may be several decades, and the development probably includes several steps. Furthermore, individual susceptibility is also important for the disease. Therefore, it has been extremely difficult to make the required observations in exposed individuals. [Pg.318]

Tocotrienols are another group of phytochemicals of rice bran which have a chemopreventive effect and have been demonstrated to inhibit breast cancer (Nesaremam et al., 1998). The polysaccharides of rice bran contain a-glucan, the anti-tumor effect of which has been demonstrated by its inhibition of gastrointestinal carcinogenesis (Akeshita et al., 1992). Rice bran agglutinin has been shown to induce apoptosis of cancer cells by the mechanism of cell cycle dysregulation (Miyoshi et al., 2001). [Pg.366]

This research is an outgrowth of an earlier project which sought to characterize the mechanism of arsenic metabolism in a population currently drinking arsenic-contaminated water in Taiwan. There were three parts to the study, a cohort study, a case control study for skin cancer, and a study of intra-individual variability associated with chronic arsenic exposure. The latter three research projects are complete and being submitted for publication. The results of this research are highly relevant since they indicate that alterations in methylation capacity may affect arsenic carcinogenesis. [Pg.268]
