Applications elution

For every protein purification problem there is always an affinity solution, but cost and safety considerations may render these solutions impractical. As an example, antibodies are widely used for analysis, where only relatively small amounts are usually required, but their production and purification on a large scale for preparative-scale chromatography may be difficult to justify economically. In some cases, Hhybridoma technology may be able to address this problem. Even if production costs are acceptable, the immobilized antibodies may be unstable over the sequence of sample application, elution, and sanitation required for multiple use of the affinity adsorbent. For these reasons, while biological ligands (antibodies, enzymes, receptors, lectin. [Pg.880]

I have carried out widespread studies on the application of a sensitive and selective preconcentration method for the determination of trace a mounts of nickel by atomic absorption spectrometry. The method is based on soi ption of Cu(II) ions on natural Analcime Zeolit column modified with a new Schiff base 5-((4-hexaoxyphenylazo)-N-(n-hexyl-aminophenyl)) Salicylaldimine and then eluted with O.IM EDTA and determination by EAAS. Various parameters such as the effect of pH, flow rate, type and minimum amount of stripping and the effects of various cationic interferences on the recovery of ions were studied in the present work. [Pg.51]

The results of sorptional properties investigation have shown the possibility of SN-SiO, application for Pb +, ClC+, Zn +, Cd + selective preconcentration. The preconcentrated Cu, Pb, Cd, Zn ions at pH 6-7 were eluted by acid and determined by atomic-absoi ption method. [Pg.274]

Equation (1) is the well-known Gaussian form of the elution curve equation and can be used as an alternative to the Poisson form in all applications of the Plate Theory. [Pg.167]

Vacancy chromatography has some quite unique properties and a number of potentially useful applications. Vacancy chromatography can be theoretically investigated using the equations derived from the plate theory for the elution of... [Pg.196]

Liquids have relatively low compressibility compared with gases and, thus, the mobile phase velocity is sensibly constant throughout the column. As a consequence, elution volumes measured at the column exit can be used to obtain retention volume data and, unless extreme accuracy is required for special applications, there is no need for the retention volume to be corrected for pressure effects. [Pg.273]

Column design involves the application of a number of specific equations (most of which have been previously derived and/or discussed) to determine the column parameters and operating conditions that will provide the analytical specifications necessary to achieve a specific separation. The characteristics of the separation will be defined by the reduced chromatogram of the particular sample of interest. First, it is necessary to calculate the efficiency required to separate the critical pair of the reduced chromatogram of the sample. This requires a knowledge of the capacity ratio of the first eluted peak of the critical pair and their separation ratio. Employing the Purnell equation (chapter 6, equation (16)). [Pg.367]

The design procedure described above will, in theory, be applicable only to samples that are separated by isocratic development. Under gradient elution conditions the (k ) value of each solute is continually changing, together with the viscosity of the... [Pg.407]

A wide range of application for small molecules through very high molecular weight polymers on H type columns has been reported since the commercialization in 1972 (34,35). Some examples of applications are summarized in Table 4.19. Table 4.19 contains the type of sample, column, and elution solvent. More applications can be found in technical literatures published by TosoHaas or Tosoh. [Pg.145]

A SEC material should be hydrophilic if it is to be used for biological applications. One such material, introduced by PolyLC in 1990 (8), is silica with a covalently attached coating of poly(2-hydroxyethyl aspartamide) the trade name is PolyHYDROXYETHYL Aspartamide (PolyHEA). This material was evaluated for SEC of polypeptides by P.C. Andrews (University of Michigan) and worked well for the purpose (Fig. 8.1). Because formic acid is a good solvent for polypeptides, Dr. Andrews tried a mobile phase of 50 mM formic acid. The result was a dramatic shift to a lower fractionation range for both Vq and V, (Fig. 8.2) to the point that V, was defined by the elution position of water. [Pg.250]

It is evident from these results that the interactive properties of the investigated SEC PS/DVB or DVB gels are very different. Because polar electroneutral macromolecules of PMMA were more retained from a nonpolar solvent (toluene) than from polar ones (THF, chloroform), we conclude that the dipol-dipol interactions were operative. Columns No. 1 and No. 2 were very interactive and can be applied successfully to LC techniques that combine exclusion and interaction (adsorption) mechanisms. These emerging techniques are LC at the critical adsorption point (18), the already mentioned LC under limiting conditions of adsorption (15,18), and LC under limiting conditions of desorption (16). In these cases, the adsorptivity of the SEC columns may even be advantageous. In most conventional SEC applications, however, the interactive properties of columns may cause important problems. In any case, interactive properties of SEC columns should be considered when applying the universal calibration, especially for medium polar and polar polymers. It is therefore advisable to check the elution properties of SEC columns before use with the... [Pg.455]

A method which uses supercritical fluid/solid phase extraction/supercritical fluid chromatography (SE/SPE/SEC) has been developed for the analysis of trace constituents in complex matrices (67). By using this technique, extraction and clean-up are accomplished in one step using unmodified SC CO2. This step is monitored by a photodiode-array detector which allows fractionation. Eigure 10.14 shows a schematic representation of the SE/SPE/SEC set-up. This system allowed selective retention of the sample matrices while eluting and depositing the analytes of interest in the cryogenic trap. Application to the analysis of pesticides from lipid sample matrices have been reported. In this case, the lipids were completely separated from the pesticides. [Pg.241]

Another application of SFC-GC was for the isolation of chrysene, a poly aromatic hydrocarbon, from a complex liquid hydrocarbon industrial sample (24). A 5 p.m octadecyl column (200 cm X 4.6 mm i.d.) was used for the preseparation, followed by GC analysis on an SE-54 column (25 m X 0.2 mm i.d., 0.33 p.m film thickness). The direct analysis of whole samples transferred from the supercritical fluid chromatograph and selective and multi-heart-cutting of a particular region as it elutes from the SFC system was demonstrated. The heart-cutting technique allows the possibility of separating a trace component from a complex mixture (Figure 12.21). [Pg.327]

Another example is the determination of bentazone in aqueous samples. Bentazone is a common medium-polar pesticide, and is an acidic compound which co-elutes with humic and/or fulvic acids. In this application, two additional boundary conditions are important. Eirst, the pH of the M-1 mobile phase should be as low as possible for processing large sample volumes, with a pH of 2.3 being about the best that one can achieve when working with alkyl-modified silicas. Secondly, modifier gradients should be avoided in order to prevent interferences caused by the continuous release of humic and/or fulvic acids from the column during the gradient (46). [Pg.346]

For preparative or semipreparative-scale enantiomer separations, the enantiose-lectivity and column saturation capacity are the critical factors determining the throughput of pure enantiomer that can be achieved. The above-described MICSPs are stable, they can be reproducibly synthesized, and they exhibit high selectivities - all of which are attractive features for such applications. However, most MICSPs have only moderate saturation capacities, and isocratic elution leads to excessive peak tailing which precludes many preparative applications. Nevertheless, with the L-PA MICSP described above, mobile phases can be chosen leading to acceptable resolution, saturation capacities and relatively short elution times also in the isocratic mode (Fig. 6-6). [Pg.164]

At the current time, there is considerable interest in the preparative applications of liquid chromatography. In order to enhance the chromatographic process, attention is now focused on the choice of the operating mode [22]. SMB offers an alternative to classical processes (batch elution chromatography) in order to minimize solvent consumption and to maximize productivity where expensive stationary phases are used. [Pg.256]
