Antithrombin III is a plasma alpha2-glycoprotem that accounts for most of the antithrombin activity in plasma and also inhibits other proteolytic enzymes. Hereditary or acquired antithrombin III deficiency results in thromboembolism. The effectiveness of treatment with antithrombin III, prepared as a concentrate from human plasma, is stiU a matter of dispute (1,2). Apart from vasodilatation, leading to a reduction in blood pressure, remarkably few adverse effects have been noted (2,3). The fall in blood pressure seems to be related to the rate of the infusion. [Pg.266]

Although there is much anecdotal evidence that TPN (as opposed to no nutrition) may be beneficial in hospitalized horses, this has not been proven (Lopes White 2002). The administration of TPN increases the risk of sepsis and thrombophlebitis in hospitalized human patients (loannides-Demos et al 1995). This may result, at least in part, from decreased intracellular killing by neutrophils (Okada et al 2000) and increased plasma thrombin-antithrombin III concentrations, which result in increased plasma coagulability (van der Poll et al 1998). [Pg.346]

Note specific activities before labeling crude bovine thrombin—96 IU/mg (Parke-Davis), pure human antithrombin III—1000 IU/mg (M. Wick-erhauzer, American Red Cross, Bethesda, MD) open implies material used as eluent in conventional chromatographic mode and ratio of loaded protein does not include antithrombin III content of defibrinated plasma. [Pg.154]

Thrombin (62 IU) loaded on the gel columns was followed by either 3 mg of crude antithrombin III or by 15 mL of 20% (w/w) bovine albumin (ALB). Key in the experiment with purified human thrombin (1072 IU), the gel columns were precoated with albumin (Hi, PVA-heparin 1, PVA and flow rate, 97 mLlh, 7 mg heparin g gel,... [Pg.157]

Classification and Prototypes Anticoagulants reduce the formation of fibrin clots. Two major types of anticoagulants are available heparin and its derivatives, which must be used parenterally and the orally active coumarin derivatives (eg, warfarin). The two groups differ in their chemistry, pharmacokinetics, and pharmacodynamics (Table 34-2). Two other anticoagulant proteins are available lepirudin, a recombinant form of hirudin, a protein found in leech saliva and human antithrombin III, a commercial preparation of an endogenous human anticoagulant. [Pg.305]

Heparin s anticlotting effect is mediated by acceleration of the action of endogenous antithrombin III, a protease that inactivates clotting factors. Patients with genetic deficiencies in antithromhin III are resistant to heparin and prone to thrombosis. Antithrombin 111 isolated from pooled human plasma is available for use in such patients. The answer is (B). [Pg.313]
