Idiotypic network. Idiotypic determinants (idiotypes) are unique antigenic epitopes characteristic of the antigen receptors on the surface of T and B cells. They are associated with the variable regions of these receptors. Antibodies produced by B cells as the result of antigenic stimulation can themselves stimulate the production of auto-anti-idiotypic antibodies which have the ability to combine with the B-cell receptor (Ig) and thus can dampen down the immune response. Idiotypes may likewise stimulate the production of T cells specific for idiotypic determinants. Jerne (1974) postulated his... [Pg.296]

In addition to monoclonal antibody or chemotherapy, immunotherapy has been developed to target tumor antigen for the treatment of FL. The unique sequence of the protein, so-called idiotype (Id) protein, can be a target of immunotherapy. Because Id protein can induce humoral and cellular immune responses against idiotype protein, vaccination with Id protein can decrease the risk of progression. However, the mechanism of anti-Id response has been unclear. [Pg.218]

Anti-idiotypic and antiergotypic Treg cells are activated after T-cell vaccination and are regulators of the immune response. The Treg cells induced by activated... [Pg.214]

Mosbach and collaborators, in 2001, described an innovative approach to drug discovery using polymers imprinted with a biologically active template [31]. The approach, called anti-idiotypic for the similarity with anti-idiotypic antibodies in the immune-response, can be used to create synthetic receptors able to generate inhibitors or receptor antagonists by exploiting the complementarity with the cavity. The imprinted cavity promotes preferentially the formation of compounds with high affinity, which can later be evaluated for the inhibitory activity and the more active selected for further analysis. [Pg.321]

A third elimination pathway occurs if anti-idiotype antibodies are formed as an immune response of the human body to the administration of mAbs. Following repeated administration, anti-idiotype antibodies are usually observed after one to two weeks, with the extent of the adverse reaction strongly depending on several factors ... [Pg.77]

Dosage regimen single doses rarely evoke a strong immunological response, whereas multiple dosing often results in anti-idiotype antibody formation. [Pg.77]

Fab (50 kd) and scFv (27 kd) antibody fragments have shorter plasma half-lives (0.5 to 21 hours) because of more rapid glomerular filtration and clearance [7], Plasma clearance, particularly of the xenogenic (usually mouse) portions of humanized and/or chimeric monoclonal antibodies, might be accelerated by development of anti-mouse (or other species), anti-isotypic, anti-idiotypic, or anti-allotypic antibodies that foster immune complex formation or reticuloendothelial clearance via several different FcR forms or elicit allergic or anaphylactic responses [7,8]. [Pg.243]
