Okadaic acid (OA), being a polyether, presents ionophoretic properties (facilitation of ion transport across membranes) as does CTX. It has been found that OA causes contraction in smooth muscles even in the absence of Ca (Ozaki and Karaki 1987 Shibata 1985). Ozaki and Karaki (1987) studied the mechanism of action of OA compared to calyculin A (another polyether isolated from a marine sponge). The results of this work suggest that OA has two separate effects activation of calcium channels as well as activating contractile elements to induce smooth muscle contraction. Recently, OA, in addition to other compounds from marine sources, has been found to be a tumor promoter that is, an agent that promotes tumor formation on already initiated cells (Fujiki 1988). It has been found that the OA class of tumor promoters bind to their own receptors which are present in particulate as well as cytosol fractions. PVC. Poly(vinyl chloride) (PVC), a very versatile polymer, is manufactured by the polymerisation of vinyl chloride monomer, a gaseous substance obtained from the reaction of ethylene with oxygen and hydrochloric acid. In its most basic form, the resin is a relatively hard material that requites the addition of other compounds, commonly plasticisers and stabilisers as well as certain other ingredients, to produce the desired physical properties for roofing use. The membranes come in both reinforced and nonreinforced constmctions, but since the 1980s the direction has been toward offering only reinforced membranes. There has been recent interest in naphtho-fused dithiepines as chiral acyl anion equivalents, particularly since the starting dithiol 128 can be obtained in enan-tiomerically pure form (89TL2575). Other effects. In addition to the compound formation and ionisation effects which have been considered, it is also necessary to take account of so-called matrix effects. FAAH was originally purified and cloned from rat liver microsomes and is able to catalyse the hydrolysis of anandamide and 2-AG, in addition to other long-chain fatty acid amides [25]. Studies into the structure and role of this enzyme have generated interest in the potential therapeutic applications of FAAH inhibitors [26-28]. FAAH knock-out mouse brains contained 15-fold higher levels of anandamide than their wild-type counterparts and these animals have also been shown to be more responsive to exogenously administered anandamide [29]. These animals also showed a reduced response to painful stimuli, supporting the hypothesis that FAAH inhibition may provide novel analgesics. Levels of 2-AG were not elevated in the FAAH knock-out animals, apparently due to the existence of alternative metabolic fates for this compound [30]. [Pg.210]
