Uptake in cells

Cheng, Z., Winant, R. C., and Gambhir, S. S. (2005). A new strategy to screen molecular imaging probe uptake in cell culture without radiolabeling using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. J. Nucl. Med. 46 878-886. [Pg.379]

Nickel (relative atomic mass = 58.69) is a first-row transition metal in group Vlllb of the periodic table. Although there are five natural isotopes, Ni (68.3%) and Ni (26.1%) are the most abundant. Proteins are often labeled with Ni (nuclear spin, I, is 3/2) to identify the nickel site by EPR spectroscopy, since paramagnetic species with significant spin on Ni exhibit a four-line splitting. Ni, which is a beta emitter with a relatively long half-life of 100 years, can be used to identify Ni-containing proteins and to measure Ni uptake in cells. [Pg.2844]

PA Porter, I Osiecka, RT Borchaidt, JA Fix, L Frost, CR Gardner. In vitro drug absorption models II salicylate, cefoxitin, a-methyl dopa, and theophylline uptake in cells and rings correlation with in vivo bioavailability. Pharm Res 2 293—298,1985. [Pg.269]

Boron analysis via detection of the prompt y-ray from the °B (n, a) Li reaction was shown to be a reliable technique (Fairchild et al. 1986). This method makes it possible to measure the boron content in small amounts (mg samples) of antibodies or boron uptake in cells grown in tissue culture. In order to determine °B concentrations in a tumor in vivo without injuring tissues, phantom prompt... [Pg.68]

Pascu et al. developed intrinsically fluorescent gallium and indium bis(thiosemicarbazonato) for rapid radiolabelling. Uptake in cells indicated localisation in mitochondria, lysosomes and additionally for indium complexes in the nucleus, therefore opening up the possibility Auger electron emission therapy via In, using a complex such as 17 (Fig. 24). [Pg.39]

The sodium-dependent multivitamin transporter is the quantitatively most important biotin transporter in the majority of human tissues. The monocarboxylate transporter 1 may play a specialized role in biotin uptake in cells from the lymphoid lineage. [Pg.185]

Some tumour growth inhibition was observed for Cu(SOD) itself [83]. The demonstrated SOD-mimetic activity (albeit low) of copper complexes such as bis(isopropylsalicylato)copper(II), prompted a study of its antitumour effect, the reasoning being that this lipid-soluble analogue would have greater uptake in cells. The complex (Structure 63) is probably of the cupric acetate type ... [Pg.156]

Regardless of the mechanism, FTIR spectroscopy shows potential as an independent modality to monitor the effects of cisplatin uptake in cells. The neural network model trained on cells from different cell lines, under different growth and instrumental conditions and tested on a independent data set, provides evidence that this could, in fact, be a tool to supplement conventional total cytosolic poly(A)-b mRNA and MTT assays. [Pg.348]

Rgure 1 shows a series of pseudo 3D distributions of DNA (x axis), BrdUrd incorporation (y axis), and cell number (z axis) obtained at hourly intervals after pulse labelling V79 Chinese hamster fibroblasts with 10 (xM BrdUrd for 20 minutes in vitro. In the profile recorded after one hour the BrdUrd labelled S phase cells are clearly identified by their green fluorescence and lie between the G1 and G2 populations. The latter two populations can also be separately identified by virtue of their lack of BrdUrd uptake and the difference in their DNA content note that the majority of cells reside in Gl. The BrdUrd labelling is classically crescent-shaped with lower levels of uptake in cells that have just entered S phase from Gl, and those found in late S phase about to enter G2. With time, the profiles show changing patterns. At two hours, the labelled cohort has become slightly skewed to the right as the cells have made more... [Pg.344]

If hepatocytes were isolated from adrenalectomized rats and placed in culture for 24 hours or less, the rate of transport by System A was enhanced when compared to uptake in cells from nonadrenalectomized animals (26). For the cultured cells from adrenalectomized rats, the stimulation of System A-mediated transport by glucagon, insulin, or dexamethasone was significantly lower than that seen in control cells (26). The decreased competence of these cells to respond to insulin and glucagon was not reversed by including dexamethasone in the incubation... [Pg.149]

Paclitaxel Folate-conjugated stearic acid-grafted chitosan oligosaccharides increased cellular uptake in cells expressing abundant folic acid receptors [27]... [Pg.145]
