Synthesis. Histamine [51-45-6] 2-(4-imidazolyl)ethylarnine (1) is formed by decarboxylation of histidine by the enzyme L-histidine decarboxylase (Fig. 1). Most histamine is stored preformed in cytoplasmic granules of mast cells and basophils. In humans mast cells are found in the loose connective tissue of all organs, especially around blood and lymphatic vessels and nerves. These cells are most abundant in the organs expressing allergic diseases the skin, respiratory tract, and gastrointestinal tract. 

In humans, the hypothalamic-derived protein and the hormone noncovalent complexes are packaged in neurosecretory granules, then migrate along axons at a rate of 1 4 mm/h until they reach the posterior pituitary where they are stored prior to release into the bloodstream by exocytosis (67). Considerable evidence suggests that posterior pituitary hormones function as neurotransmitters (68) vasopressin acts on the anterior pituitary to release adrenocorticotropic hormone [9002-60-2] (ACTH) (69) as well as on traditional target tissues such as kidneys. Both hormones promote other important central nervous system (CNS) effects (9,70).

NK cells are a subset of lymphocytes found in blood and lymphoid tissues, especially the spleen. They are about 15 an in diameter, possess a kidney-shaped nucleus and have two or three large granules in the cytoplasm. They are derived from the bone marrow. NK cells have the ability to kill certain tumour lines and normal cells infected by virus. Killing by NK cells is not specific for viral antigenic epitopes, and is not restricted by MHC molecules. They do not possess CD3 but do express CD2, CD 16 and CD56, together with a low-affinity receptor for the Fc portion of IgG.

There have been a number of observations which show increased excitation and/or reduced inhibition in slices prepared from human epileptic brain tissue. Thus burst discharges can be evoked with stimuli that would not do so in normal animal tissue and these can be blocked by NMD A receptor antagonists. The inhibitory postsynaptic currents (IPSCs) in hippocampal dentate granule cells in slices prepared from temporal lobe epileptic tissue are in fact reduced by stimulation that activates NMDA currents (Isokawa 1996), which are more prolonged than usual and show changes in slope conductance.

Heparin is biosynthesized and stored (probably as a complex with histamine and other basic species) in basophilic granules of mast cells of many animal tissues. Anandamide is found in human brain 100 pmol/g in the hippocampus, 75 pmol/g in the thalamus, 60 pmol/g in the cerebellum, and 55 pmol/g in the striatum (Martin, 1999). The concentration of AEA increases postmortem, especially when the brain is kept at ambient temperature. Furthermore, AEA surges are observed when cerebellar granule cells are treated in hypoxic conditions (Hillard, 1997). Although such concentration increases may be artifacts of postmortem brain damage, they may also occur in living tissue under certain conditions, such as hypoxia.
