Orodispersible tablet

Chewable tablets are considered to be safe for use in children with full dentition (2-3 years) (76-78) and under supervision. Orodispersible preparations in the oral cavity (e.g., tablets, films, wafers) stand also on the periphery of solids and liquids, with the extra challenge that the quantity of excipients available to improve the palatability is limited. 

Locust bean gum has been investigated for its use as superdisin-tegrant additive. An orodispersible tablet material of nimesuUde, with locust bean gum has been fabricated. It was found that a tablet formulation with 10% locust bean gum disintegrates in only 13 s (33). 

Patients preferences have been explored in a 12-week randomized, crossover, multinational, open study of 175 patients [116 ]. Most preferred olanzapine orodispersible tablets (61%) oral conventional tablets were preferred by 27%, and 12% expressed no preference. The adverse events profiles did not differ between the formulations. Mean weight increase over 6 weeks was 0.8 kg with olanzapine orodispersible tablets and 0.6 kg with conventional tablets.

Nursing staff, carers or patients may administer medicatioa Syrups and orodispersible tablets can be more acceptable and less easily cheeked or palmed than tablets. Don t crush tablets without first checking suitability with pharmacy.

Risperidone free base preparations include film-coated tablets (1, 2, 3, 4 and 6 mg), orodispersible tablets (0.5, 1, 2, 3 and 4 mg), liquids at 1 mg/mL and depot preparations. Paliperidone is available as free base tablets (3, 6 and 9 mg), the hexadecanoate (palmitate) and extended release formulation using an osmotic-controlled release oral delivery system. 

Sertindole free base appears as film-coated tablets (4, 12, 16, 20 mg) but is less commonly prescribed due to the occurrence of cardiac arrhythmias and prolongation of the QT intervals. Aripiprazole is available in a variety of preparations such as tablets (5, 10, 15 and 30 mg), orodispersible tablets (10 and 15 mg), oral solutions (1 mg/mL) and injectables (7.5 mg/mL). 

Clozapine (Fig. 18.49), a dibenzodiazepine, is used as tablets (25 and 100 mg) and appears as the free base. Olanzapine (Fig. 18.49) appears in a number of formulations including film-coated tablets (2.5-20 mg) and orodispersible tablets (5-20 mg). It can also be used as a powder for reconstitution. Zotepine (Fig. 18.49) is available as free base sugar-coated tablets (25-100 mg). Quetiapine (Fig. 18.49), on the other hand, is prepared as the fiimarate salt and tablets range between 25 and 300 mg.
