Interferon alfa was suspected of having induced the development of anti-factor VIII autoantibodies in one patient with hemophilia who survived and one without hemophilia who subsequently died from acute hemorrhage (SED-13,1094) (237,238). [Pg.1807]

In hemophilia A, the protein sequence of factor VIII is mutated so that it cannot be cleaved, resulting in a blocked intrinsic pathway. Therefore, tissue factor must be activated to stop bleeding (extrinsic pathway). Because the capillaries of joints and muscles are continually damaged by crushing when the surrounding bones and muscles move, they produce small amounts of RNA that activates the intrinsic path. Because there is no tissue factor at that site and the mutation of factor VIII in hemophilia A has blocked the intrinsic path, the affected subjects bleed into the joints. The pressure in the joint eventually stops the bleeding, and reticuloendothelial cells are recruited to remove the blood cells. The pain is relieved, but the joint structure is slowly destroyed and over time surviving individuals develop arthritis. [Pg.187]

Studies on the structure-activity relation of the metallocene dihalides were generally performed using as experimental system the Ehrlich ascites tumor (EAT) growing in female CF1 mice. The therapeutic effect was judged by determining the mean survival time (MST) and by calculating the increase in life span (ILS) compared to the untreated controls. The key-date for the survival studies was day 180 after tumor transplantation (Table I, III) or day 60 (Table VIII). [Pg.316]

Against sarcoma 180 growing as fluid tumor in the peritoneal cavity, the titanocene complexes I, VIII, XIII and XIV were tested . Best results were obtained for I which prolonged the mean survival period of mice, treated with a single injection of optimum doses, by 160-185% and caused cures of 40-50% of the animals. The other titanocene derivatives effected increases in life span of 90-140% and maximum cure rates of 20%. On the other hand, pilot studies with the ferricenium salts XLIX, LI, LIII and LV did not reveal tumor-inhibiting activity of these compounds against ascitic sarcoma 180. [Pg.126]
