CBB G-250 is less soluble than the R-250 variety. In acidic solutions it forms colloidal particles that are too large to penetrate surface gel pores and can be formulated into a staining solution that requires little or no destaining. Incorporation of the template molecule and its removal can be confirmed by the UV-vis absorption peak of the azobenzene moiety. The absorption peaks due to the azobenzene moiety at 358 nm and the Ti02-gel near 250 nm regularly increased in proportion to the adsorption cycle, which indicates that the azobenzene template is constantly incorporated into the gel film. The bound template molecule was removed by treating surface gel films with 1 wt % aqueous ammonia for 30 min. The UV absorbance at 358 nm completely disappeared upon ammonia treatment, showing complete removal of the template molecule see Fig. 6.16. [Pg.198]

The viscosities of the three hemicellulose adhesives and the starch control are given in Figure 4. The viscosity of the suspended hemicellulose adhesive was lower when compared at different solids contents than that of the starch control. It was, on the other hand, much higher than the viscosity of a raw starch suspension (30 cP at 35% solids). The higher viscosity of the raw hemicellulose adhesive is probably caused by the gel-like outer layer of the raw hemicellulose-suspended particles. The suspended starch particles showed no such surface gel effect, but remained totally undissolved. The viscosities of the cooked and raw-cooked hemicellulose adhesive were similar to those of the starch adhesive (Figure 4). [Pg.309]

For electromotive force measurements there are many suitable electrodes, and only the most novel features can be summarised here. The glass electrode is generally usable in these solvents. Within a week of dietary treatment, Harman et al. (113) have observed the formation of apical cellular vesicles in the gallbladder mucosa, PAS-staining material on the surface of the mucosa and in the crypts, and the occurrence of 1-2 mm surface gel particles staining intensely with PAS, mucicarmine, and alcian blue (see also references 5 and 8). Bile hexosamine levels were also increased (169 40 jug % versus U3 22 jug %). These findings suggest that increased mucin formation and excretion are early results of the lithogenic diet. It is not known whether this increased mucin secretion is common to all forms of cholelithiasis, what the mechanism is, or whether it is the same in both of these experimental models. In rabbits, it apparently depends on a factor excreted in bile, since it can be prevented by cystic duct ligation (116), but the factor or factors responsible have not been identified. [Pg.170]
