Stem cells additives

The potential for improved radioprotection by combining Tempol with growth factors such as stem cell factor (SGP), which protects by quite different mechanisms, has been examined in mice (82). Both SGP alone, given 20 and 4 h before and 4 h after TBI, and Tempol alone, given 10 min before TBI, increased 30-day survival, but protection was greater than additive when the two agents were combined. [Pg.490]

In addition, SDF-1/CXCR4 signaling is important in gastrulation (Nair and Schilling 2008). Thus, it is likely that CXCR4 signaling plays a central role in stem cell function from very early in development. [Pg.195]

In addition to their inherent self-sustaining properties, brain tumor stem cells may be more resistant to chemotherapy and radiation therapy than other tumor cells. Bao et al. (2006) found glioma stem cells (CD133+) were relatively radioresistant compared to CD133- tumor cells and preferentially activated the DNA damage checkpoint response. This relative resistance to standard treatment approaches of tumor stem cells compared to the majority of other cells within a tumor may underlie our current inability to cure patients with aggressive brain tumors such as glioblastoma. [Pg.257]

CFP-YFP donor-acceptor pair, YFP is several times brighter than CFP [62]. Lastly, for studying dynamic protein associations in plants, the presence of chlorophyll pigments in leaf and stem cells is an additional limitation. These pigments directly absorb the fluorescence, which decreases blue fluorescence intensity for BFP and CFP donors that can be erroneously interpreted as reduced donor fluorescence quantum yield caused by FRET [18]. If sensitized emission or FSPIM is the only available method for quantifying FRET, then it is very important to restrict measurements to chlorophyll free areas within the cells. [Pg.431]

In an attempt to mimic the physiological ratio of collagen type II to hyaluronan in the healthy human NP, Calderon et al. constructed hydrogels scaffolds composed of these two elements in a 9 1 (w/w) ratio [104]. Scaffolds were crosslinked with various concentrations of EDC/NHS, but it was found that 8 mM EDC/NHS resulted in a confined compressive modulus on the order of the native NP, while allowing for optimal rat mesenchymal stem cell (rMSC) viability and proliferation. Additionally, real time PCR results from rMSCs seeded on the scaffolds for 21 days indicated that the scaffolds promoted increased aggrecan expression and inhibited collagen type I expression compared to rMSCs cultured on monolayers. [Pg.215]

Additional future innovations likely to impact upon pharmaceutical biotechnology include the development of alternative product production systems, alternative methods of delivery and the development of engineered cell-based therapies, particularly stem cell therapy. As mentioned previously, protein-based biotechnology products produced to date are produced in either microbial... [Pg.10]

In addition to its cytotoxic effects, TNF-a appears to regulate the growth of some (non-trans-formed) cell types. It is capable of stimulating the growth of macrophages and fibroblasts, while suppressing division of haematopoietic stem cells. The systemic effects of this cytokine on cellular growth in vitro are thus complex and, as yet, only poorly appreciated. [Pg.258]

Various malignancies can also induce an anaemic state. This is often associated with decreased serum EPO levels, although iron deficiency, blood loss or tumour infiltration of the bone marrow can be complicating factors. In addition, chemotherapeutic agents administered to this patient group often adversely affect stem cell populations, thus rendering the anaemia even more severe. [Pg.278]

Mylotarg (like most other drugs) does induce some side effects, the most significant of which is immunosuppression. This is induced because certain additional (non-cancerous) white blood cell precursors also display the CD33 antigen on their surface. The immunosuppressive effect is reversed upon termination of treatment, as pluripotent haematopoietic stem cells (Chapter 10) are unaffected by the product. [Pg.386]
