The most convincing evidence in favor of a uniform 3,5-diester linkage between nucleotides has been obtained by the action of various enzymes on synthetic diesters of known constitution.218 217 Ribonuclease and spleen extracts were found to act only on nucleoside 3-(benzyl hydrogen phosphates), but not on other isomers, to give nucleoside cyclic phosphates which are broken down further to give nucleoside 3-phosphates. Since none of the compounds at the concentrations used showed a complete supression of splenomegaly, one would expect a residual viral activity in spleen extracts of mice, which received FLV suspensions preincubated with these compounds. Studies are now in progress to evaluate the leukemogenic activity of cell-free spleen extracts, prepared from mice inoculated with pretreated suspensions. Wu etal.63 have carried out such studies with RLV and rifamycin derivatives. The quality of mRNA is important in generating recombinant antibodies. It is particularly important to ensure minimal ribosomal RNA contamination when cDNA is random primed for Ubraiy construction to reduce the proportion of clones which represent non-mRNA species. The spleen mRNA of immunized mice is presmnably derived mainly from plasma cells, as the level of Ig mRNA in these cells is up to 1000-fold greater than in resting B cells. For spleens, extraction of total RNA followed ly mRNA isolation on oligo(dT)-cellulose is recommended in order to effectively process the large number of cells present in a spleen. The formation of Ru-5-P from Xu-5-P is catalyzed by another very active enzyme, phosphoketopentoepimerase (epimerase). This enzyme was first detected in spleen extracts by Ashwell and Hickman (16).
