Pharmacokinetics The absorption rate of Gonal-F following subcutaneous or intramuscular administration was found to be slower than the elimination rate. Hence the pharmacokinetics of Gonal-F are absorption rate-limited. After intravenous administration to pituitary down-regulated, healthy female volunteers, the serum profile of FSH appears to be described by a two-compartment open model with a distribution half-life of about 2 to 2.5 hours. Steady-state serum levels were reached after 4 to 5 days of daily administration. [Pg.230]

Organic acid analysis could be performed in other biological specimens [14, 22] when targeting specific analyte(s) in previously diagnosed patients, or fetuses at risk for a specific condition. Plasma/serum profiling is not helpful in the laboratory evaluation of patients without a specific diagnosis, and should be avoided [2]. [Pg.142]

HaitwickeJ al. (HIO) studied the use of high-ionic-strength solutions for the precipitation of serum proteins. In this technique a saturated solution of ammonium sulfate was added to serum and the supernatant was analyzed by RPLC after centrifugation. The recoveries for serum nucleosides and xanthine alkaloids were good, ranging from 93 to 100%. In addition, the serum profile showed no interference from this extraction technique. [Pg.20]

The use of the enzyme purine nucleoside phosphorjiase for enzymatic peak identification is illustrated by the RPLC serum profile from a patient with severe depression. Based upon the retention time, the peak eluting at approximately 15 min was tentatively identified as inosine (Fig. 14A). Cochromatography with an inosine reference compound resulted in a subsequent increase in peak area for the compound of interest (Fig. 14B). Fur-fiiermore, stopped-flow UV spectra indicated a similarity between the inosine standard and the peak tentatively identified as inosine (Fig. 15). [Pg.29]

Decreased Apo A1 appears to be a major component of the dyslipi-demic serum profile in patients with atherosclerotic occlusive disease of the lower extremities (461). Despite similar TC and HDL-C levels, A1 HDL is more anti-atherogenic than A1/A2 HDL in transgenic mice (462). This is because lecithinxholesterol acyltransferase (LCAT) and cholesteryl ester trasfer protein (CETP) are present mainly in Lp A1 and because Lp A1 A2 can inhibit Lp A1 -promoted cholesterol efflux. Also, mice overexpressing Apo A1 have more cholesterol efflux and are more protected against atherosclerosis mediated through decreased foam cell... [Pg.142]
