If a solute of interest does not contain a chromophore, it may be detected by indirect UV detection. Indirect detection is a technically simple and sensitive method for the detection of compounds with little inherent detector response. Indirect UV detection is a nondestructive technique, in which the analyte is characterized in native form. Indirect detection is a universal detection mode, with few requirements as to the exact nature of the analyte. The properties of indirect detection have been reviewed by Yeung.22 Indirect detection is particularly attractive for the analysis of biological compounds. Optical systems are the same for direct or indirect detection the only difference is that, in indirect detection, the mobile phase, rather than the analyte, contains a UV chromophore. [Pg.94]

IL-1 is produced in response to endotoxin, other cytokines, and microbial and viral agents, primarily by monocytes and macrophages. Other nucleated cells can also produce it. End Point vs Kinetic Methods. Samples may be assayed for enzymes, ie, biocatalysts, and for other substances, all of which are referred to as substrates. Mathematical models are the link between what is observed experimentally and what is thought to occur at the molecular level. Methods are described for determining the extent to which original natural color is preserved in processing and subsequent storage of foods. Color differences may be evaluated indirectly in terms of some physical characteristic of the sample or extracted fraction thereof that is largely responsible for the color characteristics. For evaluation more directly in terms of what the observer actually sees, color differences are measured by reflectance spectrophotometry and photoelectric colorimetry and expressed as differences in psychophysical indexes such as luminous reflectance and chromaticity. The reflectance spectro-photometric method provides time-constant records in research investigation on foods, while photoelectric colorimeters and reflectometers may prove useful in industrial color applications. Psychophysical notation may be converted by standard methods to the colorimetrically more descriptive terms of Munsell hue, value, and chroma.
