Proteins with molecular masses of as much as 100 kd or more can be analyzed at picomole sensitivities to give simple mass spectra corresponding to the intact molecule. This development has allowed unprecedented accuracy in the determination of protein molecular weights. [Pg.130]

Limit protein. Practice varies widely with respect to the balance between whole protein and medical food protein. Many reconunend that approximately half of the total protein allowance be given as essential amino acids and half as whole protein [25]. Others suggest that protein be limited to the WHO recommendation [26] and 20-30 % of protein requirement be given as essential amino acids [12]. In some centers, protein restriction alone (without the use of medical foods containing essential amino acids) is used [19,27]. However, protein restriction without essential amino acid supplementation may lead to chronic protein insufficiency [28]. Once the diet goals are established, the amount of medical food and the amount of standard infant formula (or breast milk) necessary to meet these goals are calculated. Medical foods for the treatment of UCD are listed in Table 15.4. They provide essential amino acids as the protein source but differ in energy, vitamin, and mineral profiles. [Pg.162]

Assessment of immunoblots may be qualitative (plus-or-minus) or more quantitative. Reliable quantitation requires concomitant analysis of control samples and of standards (usually, known quantities of the various fusion proteins). Practices such as titration of standard samples (typically, at least two orders of magnitude separated and analyzed on the same gel) are very important, as it is otherwise very difficult to evaluate differences in band intensity. [Pg.112]
