Reducing the availability of GABA by blocking the synthesising enzyme GAD also promotes convulsions. This may be achieved by substrate competition (e.g. 3-mercapto propionic acid), irreversible inhibition (e.g. allylglycine) or reducing the action or availability of its co-factor pyridoxal phosphate (e.g. various hydrazides such as semi-carbazide). Cyclization of a,j8-unsaturated acid hydrazides319 or /3-hydroxy-propionic acid hydrazides1177-1327 forms 3-pyrazolidinones. The /9-hydroxy acid derivatives have usually been cyclized with acid at elevated temperatures. If the hydrazides of aryl hydrazines are used, N-l substitution occurs. It has been reported that a,/9-unsaturated acid hydrazines cyclize by treatment of the hydrazide with nitrous acid (eq. 245),335,479,1028,1029 leading to formation of l-nitroso-... 

In a similar manner, reduction of the nitro group in the N2-(t>t/>4o-nitrophenyr)-substituted hydrazide 302 of propionic acid provides, by intramolecular cyclization of the intermediate amino compound, the l,2-dihydro-l,2,4-benzotriazine 303 followed by oxidation of the latter with potassium hexacyanoferrate to give 3-ethyl-l,2,4-benzo-ttiazine 304 (Scheme 183) <1997J(P1)3107, 2004SOS(17)357>. 

A third procedure which may be used is that due to Umberger for the determination of A -3 oxosteroids using wonicotinic acid hydrazide Full details of the method are given under Injection of Testosterone Propionate (see p. 589). 

Hydrazides have been prepared oply infrequently by the more drastic procedure of fusing the hydrazine salts of acids.- This process may give rise to secondary hydrazides in large,amount. When applied to acetic, propionic, and citronellic acids, among others, it gives good yields.
