Phosphotungstic-phosphomolybdic acid

Between the two world wars it was found that there was no need to isolate the complex acids used as precipitants they could be used in solution. The use of mixed solutions of phosphotungstic and phosphomolybdic acids was also studied, as well as the effects of changing the pH of the solutions used and of various heat treatments after precipitation on the colour of the pigments produced.

Total wine polyphenols are oxidised by the Folin-Ciocalteu reagent - composed of a mixture of phosphotungstic and phosphomolybdic acids which are reduced by the oxidation of the phenols, forming a mixture of blue oxides of tungsten and molybdenum. The blue coloration has an absorption maximum at approximately 750 nm, the intensity of which is proportional to the level of phenolic compounds present in the wine. The sequential analyser method is a direct automation of the manual method and results are expressed as a unit-less index. 

A green or blue pigment manufactured by precipitating basic dyestuffs such as malachite green or Victoria blue with solutions of phosphotungstic acid, phosphomolybdic acid, or a mixture of both.

III. The Ordinary Alkaloidal Reagents,—The reagents which precipitate alkaloids from their solutions react also with proteins. Among these may be mentioned phosphotungstic acid, phosphomolybdic acid, tannic acid, picric acid, potassium mercuric iodide, and potassium bismuth iodide. The alkaloidal reagents precipitate the majority of the proteins in acid solution only. 

Tannic acid, phosphomolybdic acid, and phosphotungstic acid precipitate peptones. Most of the acid aniline dyes and many of the organic color bases also precipitate proteins. 

White precipitates are formed when a 1% solution of cusparine is treated with phosphotungstic, phosphomolybdic or tannic acids, or with mercuric chloride or potassium mercuri-iodide. Picric acid, platinic chloride or potassium chromate give yellow precipitates, auric chloride or potassium bismuth iodide brown, and potassium ferrocyanide bluish-white (36). By treating a solution of cusparine hydrobromide with bromine water, a series of bromocusparine polybromides is obtiuned(38).

The total phenolic content of plant extracts is usually determined by colorimetric method using Folin Ciocalteu reagent. This method entails oxidation of the phenolate ion coupled to the reduction of the phosphotungstic-phosphomolybdic reagent [6, 83, 84]. The chromophore produced is a blue phosphotungstic-phosphomolybdic complex which has a maximum absorption in the region of 750 nm [22, 85]. The total phenolic content is expressed as the number of equivalents of gallic acid, tannic acid, or caffeic acid used for the calibration curves.
