The normal evoked EPSC of about 160pA obtained by focal stimulation of nearby tissue was dramatically reduced by addition of a cocktail (CABS) of CNQX 10 pM, D-APV 50 pM, bicuculline 10 pM and strychnine 5 pM to block glutamate, GABAa and glycine receptors. The small residual EPSC shown was blocked by the ATP P2 receptor antagonist suramin and is therefore probably mediated by released ATP. This variability in temporal characteristics also exists across insect species. For instance, in cockroaches certain ORNs show onset and offset of firing correlated with stimulation (Getz and Akers, 1997) whereas some ORNs in T sensilla of moths continue firing after the end of odor delivery (Anderson et al., 1995). The occurrence of excitatory and inhibitory responses in single ORNs (depolarization and hyperpolarization, respectively) was noted by Boeckh (1967) in a beetle and a blowfly and is well documented in lobsters (Ache and Zhainazarov, 1995). Inhibitory responses were also observed in patch-clamp recordings of Drosophila antennal ORNs (Dubin and Harris, 1997). Hyperpolarizing currents were due to a decrease in an outward Cl current. [Pg.672]

The eag potassium channel can be modulated by cyclic nucleotides and may play a role in ORNs. Patch-clamp recordings from antennal ORNs in eag mutants show fewer neurons responding to ethyl butyrate (Dubin et al., 1998). Application of high K+ saline to the dendritic side of ORNs raises excitability of most wild type neurons but not in eag mutants, suggesting a role for eag in transduction in ORN dendrites rather than during development (Dubin et al., 1998). [Pg.675]
