DZVP, DZVP2, TZVP DFT-optimized functions. Available for Fl(5.v) through Xe(18.vl4/ 9d) plus polarization functions. 

The SimuSolv optimize function can be used for minimization of a user defined cost expression. However, at this early stage of process development, no attempt was made to determine parameters for an economic optimum. Instead, many simulations were run to determine parameter sensitivities and malce comparisons of various possibilities which satisfied a necessary objective of reaching the desired low level of residual monomer. Food additives (like color additives) can be added at various processing stages, but the additions are commonly made at the latest production stage before final marketing to ensure optimal functionality of the additive in the product as sold. Residues of additives used at earlier production stages may be present still in the final formulation. However, if they do not have further functions at that stage, they are considered process aids (to help preparation but without technological effects in the finished product) and are not indicated on labels. In animal studies, high levels of cortisol have been shown to induce (increase) the activity of the enzyme tryptophan 2,3-dioxygenase in the liver, thereby decreasing the bioavailability of tryptophan to the brain. It is interesting to note that low acute doses of a number of different antidepressants inhibit the activity of this enzyme and, as a result, increase brain tryptophan concentrations, thus stimulating 5-HT synthesis (Badawy and Evans, 1982). In this way a link between the two key monoamine neurotransmitters and the hormone may be seen namely, reduced brain NA activity leads to decreased inhibition of the HPA axis, while increased levels of cortisol reduce 5-HT activity in the brain. Activation of the HPA axis has also been shown to result in tissue atrophy, in particular of the limbic system s hippocampus, and a reduction in the levels of neurotrophic factors responsible for the maintenance and optimal function of brain neurons (Manji et al., 2001). In conclusion, manipulation of the HPA axis (Nemeroff, 2002) and stimulation of neurotrophic factor activity (Manji et al., 2001) might open up new avenues for the treatment of affective disorders.

From a similar comparison with non-optimized functions, PEF300 (8) without and PEF400 (10) with charges, we see that the conformational details are not nearly so well reproduced. In the case of gentiobiose the optimized PEF has therefore made a real improvement.

Macromolecules, e.g., proteins, need a distinct structure within the aqueous surrounding to realize their biologic functions. This structure is stabilized inter-alia by ionic interactions between positively and negatively charged amino acid side chains and between these chains and other molecules. Optimal functionality needs a well-balanced ratio of charged residues. Each disorder of this ratio results in alterations up to complete denaturation.
