To better understand this, let s create a set of data that only contains random noise. Let s create 100 spectra of 10 wavelengths each. The absorbance value at each wavelength will be a random number selected from a gaussian distribution with a mean of 0 and a standard deviation of 1. In other words, our spectra will consist of pure, normally distributed noise. Figure SO contains plots of some of these spectra, It is difficult to draw a plot that shows each spectrum as a point in a 100-dimensional space, but we can plot the spectra in a 3-dimensional space using the absorbances at the first 3 wavelengths. The B score (Brideau et al., 2003) is a robust analog of the Z score after median polish it is more resistant to outliers and also more robust to row- and column-position related systematic errors (Table 14.1). The iterative median polish procedure followed by a smoothing algorithm over nearby plates is used to compute estimates for row and column (in addition to plate) effects that are subtracted from the measured value and then divided by the median absolute deviation (MAD) of the corrected measures to robustly standardize for the plate-to-plate variability of random noise. A similar approach uses a robust linear model to obtain robust estimates of row and column effects. After adjustment, the corrected measures are standardized by the scale estimate of the robust linear model fit to generate a Z statistic referred to as the R score (Wu, Liu, and Sui, 2008). In a related approach to detect and eliminate systematic position-dependent errors, the distribution of Z score-normalized data for each well position over a screening run or subset is fitted to a statistical model as a function of the plate the resulting trend is used to correct the data (Makarenkov et al., 2007). [Pg.249]

With a chromatographic technique capable of routinely yielding preparative fractions, quantitative and C FT NMR was the major spectroscopic tool used for chemical characterization. The established utility of and C NMR for characterization of coal products is documented well. The rank modification of von Neumann testing for data independence as described in Madansky (1988) and Bartels (1982) is applied. Although steady-state identification is not the original goal of this technique, it indicates if a time series has no time correlation and can thus be used to infer that there is only random noise added to a stationary behavior. The main goal of the data analysis is usually to find X, but the residual E can give important clues to the quality of this model. Because C NMR is less sensitive than H NMR, special techniques are needed to obtain a spectrum. If we simply operate the spectrometer in a normal (called continuous wave or CW) manner, the desired signals are very weak and become lost in the noise. When many spectra are averaged, however, the random noise tends to cancel while the desired signals are reinforced. If several spectra are taken and stored in a computer, they can be averaged and the accumulated spectrum plotted by the computer. Since the NMR technique is much less sensitive than the H NMR technique, hundreds of spectra are commonly averaged to produce a usable result.
