Nordihydroguaiaretic acid

This is an antioxidant in topical preparations. Roed-Petersen and Hjorth (1976) reported six cases of contact allergy with a lanolin cream containing nordihidro-guaiaretic acid (NDGA) as the culprit in three cases.

An intravenous infusion of PAF causes rapid (within 60 s) intravascular platelet aggregation, thrombocytopenia and platelet factor 4 release, as well as a profound and reversible neutropenia, due to enhanced aggregation and adherence of these cells. In vitro, PAF effects on neutrophils are dependent upon extracellular Ca2+ and Mg2+ and occur within 60 s of addition. One of the earliest reports of LO inhibition concerned the effects of ortho-dihydroxybenzene (catechol) derivatives on soybean 15-LO [58]. Lipophilic catechols, notably nordihydroguaiaretic acid (NDGA) (19), were more potent (10 /zM) than pyrocatechol itself. The inactivation was, under some conditions, irreversible, and was accompanied by oxidation of the phenolic compound. The orfAo-dihydroxyphenyl moiety was required for the best potency, and potency also correlated with overall lipophilicity of the inhibitor [61]. NDGA and other phenolic compounds have been shown by electron paramagnetic resonance spectroscopy to reduce the active-site iron from Fe(III) to Fe(II) [62] one-electron oxidation of the phenols occurs to yield detectable free radicals [63]. Electron-poor, less easily oxidized catechols form stable complexes with the active-site iron atom [64].

Acidity. The development of a fishy flavor in butter is well known. Cream acidities ranging from 0.20 to 0.30% appear to represent those levels at which flavor development is marginal (Parks 1974). Although the development of fishy flavors in unsalted butters is rarely encountered, it is not restricted to those products containing salt. Pont et al. (1960) induced the development of a fishy flavor in commercial butter-fat by the addition of nordihydroguaiaretic acid and citric or lactic acid. In addition, Tarassuk et al. (1959) reported the development of fishy flavors in washed cream adjusted to pH 4.6.
