I. Vinylarenes Styrene and its derivatives have been most intensively studied as substrates for asymmetric hydroformylation because of their high reactivity and high selectivity for yielding branched aldehydes. Furthermore, aldehydes derived from these olefins can be converted to various pharmaceuticals such as non-steroidal antiinflammatory agents (vide infra) [74]. Representative results for the reaction of styrene (I6a) are summarized in Scheme 7.3 and Table 7.1.

Indomethacin is mainly used as a non-steroidal antiinflammatory agent and occasionally as an analgesic clometacin on the other hand is usually recommended as an analgesic and shows weak antiinflammatory properties. Applied to serotonin, a similar reversal of a functional moiety yielded 5HT2c receptor selective agonists (Figure 15.52). 

It is responsible for almost all heart drugs, drugs for the CNS, anti-ulcer drugs, analgesics and anti-histamines. Many pharmaceuticals are made simply from readily available bulk chemicals. For example, the non-steroid antiinflammatory agent, ibuprofen, is made from toluene and propylene, two of the seven basic building blocks of the petrochemical industry. Chemical synthesis is also used to modify materials made from other sources, and chemical methods of extraction are used in the downstream processing of products from other sources.

This type of indole, with a 5-MeO group, is the basis of many pharmaceuticals—acid (48) is used to make Indomethacin, a non-steroidal antiinflammatory agent- The synthesis is straightforward if the acid is protected as an ester, HCl being used to catalyse the Fischer step.

Isoxicam is a non-steroidal antiinflammatory agent useful in the treatment of various forms of rheumatoid aurthritis, osteoarthritis and musculoskeletal disorders. It is about one-tenth as potent as its structural relative svidoxicam its similar long Tlj (>30 hrs.) adlows once-daily dosing. 

Oxaprozin is a non-steroidal antiinflammatory agent indicated for use in various forms of rheumatoid arthritis, osteoeirthritis, and ankylosing spondylitis. A long Th allows once/twice daily dosing. Structur ly it is somewhat unique, being a 3-substituted, rather than a 2-substituted propionic acid.
