New modes of action

The development of new chemotherapeutic agents for a particular disease state continues to depend to a significant degree on the discovery of a lead compound , which can be further developed into a viable drug candidate. A very important aspect of this process is to find novel substances that will provide a new mode of action for the treatment of a specific disease. From a historical standpoint, the isolation of novel substances from natural sources has often provided this lead compound. The number of new drugs and their origin for the period 1981-2002 has recently been summarized [1], and, of 79... 

In the search for bioactive marine secondary metabolites, drugs or drug leads with new modes of action, many invertebrates with new modes of action have been explored and tested in a variety of assays. Many marine natural products were found to be cytotoxic to an assortment of tumor cells, Some parts of the marine natural products were also found to be ichthyotoxic so as to defend themselves. Two examples, out of many, are the latrunculins and sarcophine. ... 

Natural products represent an immense reservoir of new molecules for the development of pesticide agents. In particular, natural products based herbicides have become attractive in recent years for a few reasons. One of the reasons is that they are environmentally benign. Furthermore, the novelty of their structures offers the possibility of finding compounds with new mode of actions, thus diminishing the induction of plant resistance, a problem associated with commonly used synthetic herbicides. 

Most of the top 25 companies are involved in discovery research targeted at the synthesis of new chemicals with new chemical structures, new modes of action and low rates of application that can be protected by international patents and that will give the inventing company an advantage over its competitors. Other generic manufacturers also invest in research but this is more applied in terms of manufacturing opportunities and formulation advances. 

In addition, agrochemical companies continue to search for new compounds with novel chemistry and with new modes of action that can be incorporated into crop protection management strategies that will control resistant insects. Furthermore, there is also the opportunity to introduce synergists of insecticides that depress or inhibit the detoxifying enzymes and, thereby, enhance the effectiveness of the compounds.

They can target the ongoing demand for modem crop protection compounds by introducing new products with new modes of action and higher efficacy. 

New agrochemicals introduced in the past five years include new chemistries with known modes of action, such as the protoporphyrinogen inhibitor bencarba-zone, the phytoene desaturase picolinafen and beflutamid, and sodium channel pyrethroids new chemistries with possibly new modes of action, such as flonic-amid and pyridalyl and new chemistries with established new modes of actions, such as flubendiamide, which activates ryanodine-sensitive intracellular calcium release channels, ryanodine receptors RyR, in insects. 

Knowledge of pest biochemistry will open multiple target sites to which agrochemicals of appropriate structure could be directed. This is to say that new modes of action will emerge and may overcome some of the presently unmet needs. These include such items as ...

Most importantly, the chart indicates how two classes with new modes of action, the neonicotinoids and strobilurins, both introduced in the last decade, continue to stimulate the market, suggesting that a focus on new modes of action remains an excellent recipe for success.

Improvements include increased effectiveness, fewer side effects, a new mode of action, or a new drug form, for example, pills versus injections, that will improve patient compliance and quality of life. 

Mirtazapine (Remeron) is a new antidepressant with an entirely new mode of action. It is an alpha2-adrenoreceptor blocker, producing increased noradrenergic and serotonergic activity. Common side effects include sedation and nausea. 

Many challenges are encountered on the path to conversion of a drug lead with an attractive new mode of action into a clinically useful therapeutic agent. As well appreciated by drug hunters, more risk is associated vdth the development of an NC E than an improved representative from an already widely used and proven class of antibiotics (with the combination of clinical benefits and appropriate toxicological... 

On the other hand, a new mode of action is sometimes discovered for an existing substance. If further substances are developed with a similar mode of action, the question arises whether a new stem is needed, which would mean modifying the old name for the first compound in the series. For example, albifylline and pentoxifylline are N-methylxanthine derivatives and the stem -fylline was therefore chosen for their names. These substances have been found to also suppress tumor necrosis factor-a. The experts decided to retain the stem fylline in this case, since the new action was nevertheless based on the typical xanthine-mediated inhibition of phosphodiesterase. 

Pseudomonic acid A can be hydrolysed to monic acid A, which can then be converted into a variety of semi-synthetic esters and amides, many of which are herbicidal because they inhibit ITRS in plants. As noted above, this was the first time that herbicidal inhibitors of this enzyme had been discovered, so this important observation validated a new mode of action for herbicides. F. Matsumura There are certainly other such examples. One that comes to mind is the carbamate insecticides which came originally from a natural product model. Although I think it is true that every 10 or 20 years someone will discover a completely new structure with a new mode of action such as Merck has done with the avermectins, there may come a time when we run out of really good models for synthesis. Then we may want to come back to some of the older but proven targets such as GABA receptors for the design of new insecticides. 

VIII. Discover and Develop Fungicides with New Modes of Action. The discovery of new fungicide target sites is required to compensate for the loss of products due to de-registration and resistance.
