Mucin preparation

It is of interest to note that we have obtained the same fully methylated L-fucose derivative from Morgan s hog mucin preparation (see p. 44) and it is indeed probable that the group substances from pepsin and hog mucin are identical in many details of their structure. 

Mice and rats, rendered anemic by phenylhydrazine poisoning, were given oral mucin preparations from hog gastric mucosa, which significantly accelerated hematological remission (C27). This effect was related to a modification of intestinal flora or to an improvement of intestinal absorption of iron or vitamin B12. 

Piccinini reported a nitrogen retention-promoting effect in normal and hypophysectomized rats of hog mucin rich in blood oup substances (P7, P8). Cresseri et al. noted retention of amino acid nitrogen in rats treated parenterally with mucin preparations and vitamin B12 (C27). 

This was discovered in regard to various Streptococcus and Neisseria species by Smith (S25) with the use of a hog mucin preparation. Smith etal. (S25-S27) and Record and Grinstead (R2, R2a) then demonstrated that this activity was not due to a single factor, but to a syner c combination of three factors a viscous medium, a nonspecific particulate matter, and an important third factor. The latter consists of (1) heparin, (2) chondroitin sulfuric acid, and (3) neutral blood group mucoid.

For many years a solution of crude acetone powder of bovine or porcine pancreas has been used to disaggregate tissues and to release cultured cells from their substratum. These preparations — referred to as trypsin 1 250 (based on an international standard) — contain not only trypsin but a range of enzymes including chymotrypsin and elastase which are equally important. Purified trypsin seldom is as efficient as the cruder preparations, especially on tissue disaggregation, when mucinous clumps result (Ronaldoni, 1959).
