Microarrays gold-coated

For microarray work, PLL-PEG-biotin-sAV monolayers adsorbed onto underlying titanium dioxide surfaces (pillars) replaced the b-SAM gold-coated glass surface. While oriented capture agents out-performed their random counterparts, distinct differences in Fab and antibody performance between this new surface and the b-SAM surface were apparent. 

Many instruments for contact or non-contact microarray printing are now commercially available. We have been using the Micro Array 03A (Wittech Co., Ltd.) to print our microarrays. This machine uses flat-tip gold-coated needles the needle tip diameter used for our experiments ranges from 100 pm to 500 pm. 

One further development of this approach has been to link chelating iminodiacetic acid groups to PEG, which in turn can be bound by Cu ions and so provide a highly specific binding site for Hisj-tagged proteins (Cha et al., 2004). Another variation is gold-coated microarrays, which have the advantage that they can be combined with surface plasma resonance (SPR) and MS for further detection and analysis of the captured molecules (Bieri et al., 1999 Houseman et...

An interdigitated gold microarray electrode coated with Nation has recently been used to detect serotonin in human whole blood samples (20 pL) to monitor allergic response, during which serotonin is secreted into the bloodstream [111]. Results showed excellent agreement between electrochemically monitored serotonin... 

Other hydrophihc polymers have been conceived as alternatives to dextran [6]. For example, polyvinyl alcohol and polyacryl acid derivatives are feasible and graft combinations thereof have been shown to be applicable to SPR detection [14]. Poly-L-lysine has become popular for DNA microarray coatings, due to its highly positive charge. It has also been attached to SAM-derivatized gold surfaces for subsequent modification with thiol reactive groups [15].
