The separation of mixtures involving N-methyl-JLtetrahydropyridines into their pure components by means of gas-liquid chromatography was discussed in a report by Holik et al. (87). They found that, using tris(/3-cyanoethoxymethyl)-y-picoline as the stationary phase, the primary factors involved in the specific retention volumes of these enamines is the electronic effect of a methyl substituent and the nitrogen atom on the carbon-carbon double bond. The extracted fractions were esterified with either BF3-MeOH reagent or diazomethane and analyzed by GLC. Gas liquid chromatography (GLC) was conducted with a Perkin-Elmer Sigma 3 equipped with flame ionization detector. Separations were obtained on a Hewlett Packard 12 m x 0.2 mm i.d. capillary column coated with methyl silicon fluid (OV-101). The temperature was maintained at 80°C for 2 min then programmed from 80 to 220°C at 8°C/min. The injector temperature was 250°C. Mass spectra were obtained on a Hewlett Packard model 5995 GC-MS mass spectrometer, equipped with a 15 m fused silica capillary column coated with 5% phenyl methyl silicone fluid. Spectra were obtained for major peaks in the sample and compared with a library of spectra of authentic compounds. 

Analysis Techniques. The contents of the major breakdown products of xetralin (naphthalene and 1-methyl indan) present in the distillate were determined by gas-liquid chromatography using a Hewlett Packard Series 5750 Research Chromatograph with a 62m x 0.5mm diameter glass capillary SCOT column coated with nonpolar SE 30 liquid phase (see Reference (4 ) for details). 

Haworth methylation of methyl /3-D-glucopyranoside and its 4-benzyl and 4-(tetrahydropyran-2-yl) ethers was investigated in connection with partial-methylation studies on cellulose.267 For the unsubstituted glycoside, the ratios of relative rate-constants k2 k3 k4 k were estimated to be 8 2 1 8, and, for the 4-ethers, it was found that ke> k2> k3 best agreements between calculated and experimental yields were found with the assumption that the rate constant for reaction at HO-3 is doubled when HO-2 is substituted. Later methylation studies,268 performed to low degrees of substitution, with analysis by gas-liquid chromatography, gave k2> k4> k3 for the reactivity...

The structure of monomorine I (16a), isolated from the extract of workers of the thief ant, Monomorium pharaonis, by gas-liquid chromatography (Table II), has been revealed to be 3-butyl-5-methyloctahydroindolizidine on the basis of mass and H-NMR spectra. The mass spectmm shows characteristic peaks at m/z 180 (345 M - CH3) and m/z 138 (346 M - Bu, a base peak), indicating the presence of methyl and butyl group attached to the a carbons of an amine, in addition to a molecular ion at m/z 195. The fact that the (M — 1) peak is higher than the M peak indicates a cyclic amine. The H-NMR spectmm shows two methyl groups (triplet at 8 0.88, CH3—CH2 doublet at 8 1.18, CH3CH—N) and three methines adjacent to nitrogen (8 2.13, 2.27, and 2.52).

The starting material for the synthesis of the 4-substituted derivatives 176 was the tautomeric 4(7)-nitrobenzo-triazole 173, which upon methylation with dimethyl sulfate in aq NaOH afforded the 4-nitro-2-methylbenzotriazole 174. The H NMR spectrum of the purified reaction mtKture after methylation showed the existence of all three triazole ring iV-methylated isomers in equal amounts. Compound 174 was isolated by virtue of its insolubility in cone HCl. Purity of products was confirmed by gas liquid chromatography (GLC). 

In examining the structure of a polysaccharide, it is convenient to consider the methods involved under the three main headings (a) quantitative analysis, (b) methylation, and (c) periodate oxidation. These techniques may be supplemented by partial or enzymic hydrolysis as the circumstances indicate. Each of these aspects of polysaccharide chemistry may be aided by the application of gas-liquid chromatography, either qualitative or quantitative, or both. Thus, separations impossible by other techniques may often be achieved, and analytical data obtained in a fraction of the time demanded by other methods. 

The present article is concerned with the use of gas-liquid chromatography for neutral, basic, and acidic sugars and some of their simple derivatives. A succeeding article will treat the separation of methylated sugars and their derivatives. 

The analysis of monosaccharide mixtures as the permethylated derivatives was proposed early in the application of gas-liquid chromatography to carbohydrates, but the method has now been superseded by more convenient procedures.230,231 There are, however, situations in which this method is useful, such as during a structural study of a polysaccharide by the methylation technique. The mixture of partially methylated monosaccharides obtained by methanolysis may then be fully methylated, and the proportions of the various monosaccharides determined. This approach has been used, for example, in studies on a galactomannan392 and on tamarind-kernel polysaccharide.393 Such an analysis also constitutes a useful check to ensure that no significant change in the composition of the polysaccharide occurred during methylation. 

In the carbohydrate series, the acids encountered are aldonic, aldaric, uronic, or saccharinic acids. Often, these acids are readily transformed into their lactones or methyl esters, and it is as derivatives of these that they are commonly studied. Table VIII (see p. 136) records examples in which derivatives of such acids and lactones have been subjected to gas-liquid chromatography. 

The occurrence of 3-deoxy-D-manno-octulosonic acid in lipopoly-saccharides has prompted its synthesis,537 together with the D-gal-octo158 and the D-gluco analogs.157 Although gas-liquid chromatography was successfully used to analyze the products of these syntheses, it has been reported that methanolysis of a bacterial endotoxin lipopolysaccharide failed to yield 3-deoxy-D-manno-octulosonic acid, presumably because of the lability of the latter to acid.381 However, Kasai and Nowotny have reported four peaks for the O-trimethylsilyl derivatives of 3-deoxy-D-manno-octulosonic acid obtained by methanolysis of the glycolipid from a Salmonella minnesota mutant.538 Reduced 3-deoxy-D-manno-octulosonic acid and its methyl ester have also been analyzed successfully as their acetates.339,539...
